306-O12B-3 is an ionizable cationic lipidoid.1 It has been used in the generation of lipid nanoparticles (LNPs) for the delivery of antisense oligonucleotides (ASOs). LNPs containing 306-O12B-3 localize to the liver after intravenous administration in mice. LNPs containing 306-O12B-3 in combination with other lipids encapsulating ASOs targeting the gene encoding proprotein convertase subtilisin kexin type 9 (PCSK9) decrease hepatic PCSK9 expression in mice. Intravenous administration of LNPs containing 306-O12B-3 and the cationic lipidoid NT1-O14B encapsulating ASOs against tau decreases tau expression and levels in mouse brain.2
