Amylin is a 37-residue peptide hormone secreted from pancreatic β-cells that reduces food intake, decreases glucagon secretion, slows gastric emptying, and increases satiety. It binds to amylin receptors with IC50 values of 2 and 0.2 nM for porcine and guinea pig nucleus accumbens membrane preparations, respectively. Amylin is present in amyloid aggregates derived from the pancreatic islets of humans with type 2 diabetes. Amylin forms fibrils that react to thioflavin T and are organized in a cross-β arrangement of β-strands similar to amyloid-β (1-40) and amyloid-β (1-42) fibrils., Aggregated amylin (60 μM) decreases the viability of primary pancreatic islet cell cultures by 60%. It also inhibits electrically-stimulated contractions of guinea pig vas deferens (EC50 = 213 nM).
