TUNEL Assay Kit: Advanced Detection of Apoptotic DNA Fragmentation

Hello everyone, Mr. G brings us new experimental knowledge, TUNEL method for detecting cell apoptosis. Below, I will provide a systematic introduction to the experimental principles and application scenarios.

1. Tunel method for detecting apoptosis principle

During the middle or late stages of apoptosis, some specific DNA endonucleases are activated, which cleave genomic DNA into multimers of 180~200bp fragments between nucleosomes, exposing 3'-OH ends at each DNA fragments terminal. While in normal or proliferating cells, there is almost no DNA fragmentation, resulting in minimal formation of 3'-OH ends. TdT enzyme (Terminal deoxynucleotidyl transferase) catalyze fluorescent-marked dUTP label to the 3'-OH ends of fragmented DNA, which can be detected with fluorescence microscope.
