1-Oleoyl-Lysophosphatidsäure (LPA) ist ein natürlich vorkommendes Phospholipid mit verschiedenen Wirkungen. LPA ist in vielen Geweben weit verbreitet. Ähnlich wie viele andere Biomediator interagiert LPA über spezifische Zelloberflächenrezeptoren, wie G-Protein-gekoppelte Rezeptoren, um biologische Effekte zu induzieren.
In vitro: LPA könnte die Signaltransduktion über den Edg/LPA-Rezeptor vermitteln und dadurch die Proliferation, Migration, Adhäsion und antiapoptotische Funktion von Tumorzellen induzieren. Darüber hinaus wurde festgestellt, dass LPA eine Hochregulation von VEGF auslösen kann, was zu einem indirekten Einfluss auf Initiation und Progression von Malignitäten führt. LPA könnte auch die Sekretion von Matrix-Metalloproteinase und Tumor-Angiogenese-Faktor stimulieren [1].
In vivo: LPA wurde auf seine Wirkung bei der vaskulären Remodellierung in einem Rattenmodell für hypoxische pulmonale Hypertonie getestet. Das Serum von Tieren in der hypoxischen Gruppe zeigte höhere chemotaktische Eigenschaften gegenüber primären Lungenfibroblasten von Ratten, und diese Zunahme der Zellmigration wurde durch die Antagonisten des LPA-Rezeptors 1 und 3 verhindert. Es stellte sich auch heraus, dass LPA die adhäsiven Eigenschaften von menschlichen Endothelzellen der Pulmonalarterie erhöht, indem es den LPA-Rezeptor 1 oder 3 aktiviert [1].
Klinische Studie: Klinische Ergebnisse zeigten, dass die LPA-Konzentration bei Patienten mit Bauchspeicheldrüsenkrebs signifikant höher war. Zur Diagnose von Bauchspeicheldrüsenkrebs betrug die Sensitivität von LPA 89,6% und die Spezifität 79,4%. Allerdings hatte eine Veränderung des Plasmalipidprofils in Bezug auf LPA signifikante Korrelationen mit der Tumorgröße, dem pathologischen Stadium, den umgebenden Lymphknoten sowie spezifischen histopathologischen Merkmalen [2].
