K252a ist ein Staurosporin-Analogon, das aus Bodenpilzen der Art Nocardiopsis sp. isoliert wurde und Protein-Kinasen (PK) C, PKA, Ca2+/Calmodulin-abhängige Kinase Typ II und Phosphorylase-Kinase mit IC50-Werten von 470, 140, 270 bzw. 1,7 nM hemmt.[1],[2] Da es Neurotrophin-Rezeptor-Tyrosinkinasen hemmt, wird K252a in Konzentrationen von 100-500 nM eingesetzt, um die Proliferation von Trophoblasten zu unterdrücken und Apoptose zu erhöhen; dies geht mit einer Störung der mitochondrialen Funktion in kultivierten Choriokarzinomzellen einher.[3] Kürzlich wurde gezeigt, dass K252a PRK1 (IC50 = 3.2 nM in vitro), eine PKC-verwandte Kinase inhibiert; diese phosphoryliert Histone H3 an Threonin-11 und ist an der androgenabhängigen Genexpression beteiligt.[4]
