Background

Der Ubiquitin-Proteasom-Weg spielt eine wichtige Rolle bei der selektiven Degradation von intrazellulären Proteinen. Neben der Beseitigung beschädigter oder fehlgefalteter Proteine reguliert dieser proteolytische Weg auch die Verfügbarkeit wichtiger Proteine, die an der Kontrolle von Entzündungsprozessen, Zellzyklusregulation und Genexpression beteiligt sind. (R)-MG132 ist ein potenter, reversibler und zellpermeabler Proteasominhibitor. Nach einer Behandlung für eine Stunde mit 100 nM hemmt es 50% bzw. 31% der Proteasomaktivität in Lysaten von J558L-Multiple-Myelom-Zellen bzw. EMT6-Brustkrebszellen. Das Stereoisomer (R)-MG132 ist ein effektiverer Inhibitor für chymotrypsinähnliche (ChTL), trypsinähnliche (TL) und peptidylglutamylpeptid-hydrolysierende-Proteasome(PGPH)-Aktivitäten im Vergleich zu (S)-MG132(IC50s = 0,22 versus 0,89 μM(ChTL);34,4 versus104,43μM(TL);2,95versus5,70μM(PGPH)).
