Description of 2-Methoxyestradiol (2-MeOE2)

2-Methoxyestradiol (2-MeOE2) est un inhibiteur de HIF-1α. Il peut inhiber l'accumulation de HIF-1α dans les noyaux ainsi que l'activité transcriptionnelle de HIF[1-3]. Le 2-ME peut déclencher l'apoptose induite par p53 par deux mécanismes : l'activation de P38 et NF-κB ainsi que l'activation de JNK et AP-1, conduisant à la phosphorylation de Bcl-2. Il empêche la mitose des cellules tumorales en inhibant l'assemblage de la tubuline, en déstabilisant les microtubules et en interférant avec la mitose du fuseau. En tant qu'inducteur d'apoptose et inhibiteur de l'angiogenèse, il présente une activité antinéoplasique potentielle[4].

Le traitement au 2-Méthoxyestradiol (10 µg/mL ; 1 à 4 jours) a significativement inhibé la capacité de prolifération des cellules de mélanome B16[5]. Le 2-Méthoxyestradiol (0,03-33 mM) a supprimé la migration et la prolifération induites par le L-lactate dans les cellules d'ostéosarcome[6].

Le traitement au 2-Méthoxyestradiol (30 mg/kg ; i.p./p.o ; 7 jours/3 semaines) avant et après irradiation a inhibé les lésions cutanées induites par irradiation (RISI) chez les souris C57/BL6. Le 2-ME a également réduit l'inflammation induite par irradiation, l'épaisseur de la peau et la fibrose vasculaire[7].

Méthode de préparation

Les cellules B16 ont été ensemencées dans des plaques de 96 puits (1000 cellules par puits) et traitées avec 2-Méthoxyestradiol (10 µg/mL) après l'adhésion cellulaire. Le test CCK8 a été réalisé aux jours 1, 2, 3 et 4 après le traitement.

Le traitement avec 2-Méthoxyestradiol a significativement inhibé la capacité de prolifération des cellules de mélanome B16.

Méthode de préparation

La peau des souris a été irradiée avec 17 et 20 Gy à l’aide d’un champ de 4 cm. Le 2-Méthoxyestradiol a été injecté dans le groupe d’irradiation.
