Preparation Method

The conditioned media were replaced with new media containing AS-IV at the final concentration indicated with or without 200 µM of 6-Hydroxydopamine hydrobromide.

Applications

Compared with vehicle controls, exposure to 200 µM 6-Hydroxydopamine resulted in the loss of 75% of PNCs, as determined using the LDH assay.

Preparation Method

The animals received a single, intrastriatal injection of 6-Hydroxydopamine hydrobromide. Twenty-five micrograms of 6-Hydroxydopamine hydrobromide, which was dissolved in 1.5 µl of normal saline with 0.2% ascorbic acid, was injected at a depth of 5.6 mm ventral to the skull at the same anterior/posterior and medial/lateral coordinates at which the cells had been injected. The injection site was chosen to be midway between the two implant sites. 6-Hydroxydopamine hydrobromide was injected over 5 min, and the needle was left in place for an additional 5 min before withdrawal of the needle.

Background

Le chlorhydrate de 6-hydroxydopamine (6-OHDA) est un analogue structural des catécholamines, la dopamine et la noradrénaline, et exerce ses effets toxiques sur les neurones catécholaminergiques. La neurotoxine 6-hydroxydopamine continue de constituer un outil topique précieux utilisé principalement dans la modélisation de la maladie de Parkinson (PD) chez le rat [1].

La méthode classique d'infusion intracérébrale d'hydrobromure de 6-hydroxydopamine, impliquant une destruction massive des neurones dopaminergiques nigrostriataux, est largement utilisée pour étudier les dysfonctionnements moteurs et biochimiques dans la maladie de Parkinson [1].

Le chlorhydrate de 6-hydroxydopamine subit une auto-oxydation robuste générant du H2O2 cytotoxique, des espèces réactives de l'oxygène (ROS) et des quinones de catécholamines qui attaquent les groupes nucléophiles endocellulaires [2].

Les effets neurotoxiques de l'hydrobromure de 6-hydroxydopamine se produisent par un mécanisme en deux étapes impliquant l'accumulation de la toxine dans les neurones catécholaminergiques, suivie d'une altération de l'homéostasie cellulaire et des dommages neuronaux. Le stockage intracellulaire de l'hydrobromure de 6-hydroxydopamine est médié par les transporteurs membranaires de dopamine ou noradrénaline (DAT et NAT respectivement), qui reconnaissent et absorbent l'hydrobromure de 6-hydroxydopamine en raison de sa similitude structurelle avec les catécholamines endogènes [3]. L'hydrobromure de 6-hydroxydopamine est infusé unilatéralement dans le MFB, produisant un déséquilibre fonctionnel entre les systèmes nigrostriataux dopaminergiques et entraînant une lenteur motrice, indicative d'une akinésie parkinsonienne-like, ainsi qu'un comportement rotationnel typique en réponse aux agents dopaminomimétiques [4].
