Description of Coelenterazine 400a

Coelenterazine 400a, un dérivé de coelenterazine naturel[1], sert d'un substrat de luciférase de Renilla( Rluc)[2], avec la longeur maximale d'onde d'émission de 400nm.En présence de Rluc, Coelenterazine 400a réagit avec l'oxygène moléculaire, se forme des intermédiaires à haute énergie et émet de la lumière en conséquence[3][4]. Ses interférence minimal aux accepteurs fluorescents le fait devenir un choix idéal pour les essais de transfert d'énergie par résonance de bioluminescence(BRET), c'est communément utilisé pour évaluer les interations de protéine-protéine[3][4].

Protocol of Coelenterazine 400a

Procotole expérimentale de Coelenterazine 400a dans l'essai BRET
1. Préparation de solution de stock: Dissoudrez Coelenterazine 400a dans l'éthanol à 100% ou le méthanol pour préparer une solution de stock de concentration de 1nM. Prendrez une aliquote de la solution de stock et stocker la solution de stock à -20°C ou -80°C en obscurité.
2. Préparation d'échantillon: Cultivez les cellules dans un milieu appriprié jusqu'à ce qu'elles atteignent la confluence de 70-80%. Utilisez les réactifs de transfections approriés pour le co-transfert des cellules avec la luciférase de Renilla et les plasmides d'accepteur fluorescent. Après la transfection, continuez cultiviter les cellules pendant 24-48 heures pour assurer l'expression adéquate de protéine.
3. Préparation de solution de substrat: Avant l'utilisation, diluez la solution de stock de Coelenterazine 400a avec le milieu de culture cellulaire jusqu'à ce que la concentration de solution atteint 100M.
4.Essai BRET:Lavâtes les cellules transférées et les remettrez dans le phosphate tamponné saline(PBS).Ajoutez la suspension cellulaire de 90µL dans chaque puits de la microplaque blanche à 96 puits. Ajoutez la solution de substrat de 10µL dans chaque puits pour achever la concentration de 10µL finale .
5.Mesure de fluorescence:Après ajout de Coelenterazine 400a, mesurez l’intensité d la luminescence à l’aide du lecteur de microplaque multimode.
