Le ferrozine réagit avec le fer divalent pour former une espèce complexe magenta stable et est utilisé pour la détermination directe du fer dans l'eau avec une absorbance maximale à 562 nm [1,2]. Le spectre d'absorption visible du complexe ferreux de ferrozine présente un pic unique et net à 562 nm. À cette longueur d'onde, l'absorptivité molaire est de 27 900 et la loi de Beer-Lambert est respectée jusqu'à environ 4 mg/L de Fe [1].
Test de Ferrozine sur les cellules ISE6.
Le test à la ferrozine pour mesurer le fer non héminique a été adapté afin de déterminer la concentration en fer dans les cellules ISE6. Après une diminution et/ou exposition au fer, les cellules ont été collectées et les lysats cellulaires ont été obtenus en utilisant la méthode décrite ci-dessus. De l'acide chlorhydrique concentré (99,5 %) a été ajouté puis chauffé à 95 °C. Après refroidissement à température ambiante, le mélange a été centrifugé et le surnageant obtenu auquel on a ajouté 75 mM d'ascorbate ou de l'eau. Ensuite, 10 mM de ferrozine ont été ajoutés. De l'acétate d'ammonium saturé a été ajouté pour faciliter le développement de couleur. L'absorbance a été mesurée à 550 nm et la concentration en fer calculée sur base du coefficient d'extinction molaire du complexe fer-ferrozine qui est de 27 900 M-1cm-1 ainsi que sur base de la concentration protéique qui elle-même avait été mesurée grâce au kit Micro BCA Protein Assay Kit. La concentration totale en fer est calculée à partir des échantillons avec ascorbate tandis que celle en ferreux est calculée à partir des échantillons sans ascorbate (agent réducteur). La concentration en ferrique quant à elle est obtenue par différence entre la concentration totale en fer et celle en ion Fe2+.
