Preparation Method |
The C-terminal end of S. pombe GT was mutated from PDEL to the S. cerevisiae retrieval signal HDEL, and the GT-encoding gene, gpt1+, was then inserted in high-copy number expression vectors (p426GPD) under the glyceraldehyde 3-P dehydrogenase promoter. S. cerevisiae alg6 cells (PRY103) were then transformed with the expression vectors with or without S. pombe gpt1+ gene. |

Uridine diphosphate glucose helps with the synthesis of b-1,6-glucan, one of Saccharomyces cerevisiae cell wall components, in the lumen of the endoplasmic reticulum (ER). |

Mice of various strains, Balb/c, CD8, and CRF 1with and without tumor implants, maintained on Purina rat chow and tap water, were infused intraperitoneally with a phosphate-buffered saline (PBS) solution of Uridine diphosphate glucose at a constant rate of 0.8 ml/day for 5 days. |

The effects of Uridine diphosphate glucose on the levels of 5-phosphoribosyl pyrophosphate (PRPP) and PRPP synthetase in mouse liver, spleen and transplanted tumors are investigated to determine the mechanism of action with three stains of mice models. |

L'uridine diphosphate glucose (glucose-uracile-diphosphate, UDPG) est un sucre nucléotidique. Il est utilisé dans le métabolisme des sucres nucléotidiques en tant que forme activée du glucose, un substrat pour les enzymes appelées glucosyltransférases. [1] L'uridine diphosphate glucose a montré des effets spécifiques aux tissus qui se sont avérés être utiles dans le traitement de certaines maladies du foie. On sait également qu'il a plusieurs effets sur le métabolisme intra-hépatique de la bilirubine qui, à leur tour, sont liés à la synthèse de glycogène se produisant dans le foie.[2]
Une étude in vitro a démontré que les effets de l'UDPG sur le métabolisme cellulaire ne semblent pas se limiter à l'induction enzymatique. D'autres ont rapporté que l'UDPG peut avoir des effets dans des conditions qui empêchent l'induction enzymatique. Les résultats ont montré qu'une quantité significative d'UDPG, bien qu'il s'agisse d'un composé hautement polaire, passe à travers la membrane sans être modifié. Une grande fraction de l'UDPG ajoutée au milieu d'incubation a cependant été trouvée dans la cellule sous forme de glucose phosphate, indiquant une clivage après pénétration dans la cellule. Finalement, tout l'UDPG entrant dans les cellules était dégradé en glucose phosphate et glucose. Les études in vitro montrent que le G-6-P n'est pas la forme active et plusieurs tests montrent que l'uridine ne conduit pas à des changements similaires à ceux observés avec UDPG.
Une étude in vivo a indiqué que le niveau intracellulaire de PRPP dans les tissus animaux est grandement affecté par l'UDPG extracellulaire. L'altération du niveau de PRPP par UDPG n'est pas linéairement dose-dépendante. Les changements induits par UDPG dans la PRPP étaient également spécifiques aux tissus : le foie de souris était plus sensible que la rate. Cette dernière spécificité pourrait bien être liée aux effets thérapeutiquement bénéfiques de l'UDPG.[2]
