Description of GS967

GS967 est un inhibiteur puissant et sélectif du courant sodique tardif (late INa) dans les myocytes ventriculaires et les coeurs isolés, ses valeurs d'IC50 sont respectivement 0,13 et 0,21µM[1]. GS967 peut induire un décalage hyperpolarisé de l'inactivation stable du canal, ralentir à la fois la récupération après l'inactivation rapide et l'apparition de l'inactivation lente[2]. GS967 a été largement utilisé dans divers modèles pour bloquer les canaux sodiques cardiaques, exerçant des effets antiarythmiques[3].

Le traitement in vitro avec GS967 a inhibé le courant sodique de pic (INaP) avec un blocage dépendant de l'usage (BDU) dans les cardiomyocytes dérivés de cellules souches pluripotentes en 400 millisecondes, sa valeur d'IC50 est 0,07µM[4].

Le traitement in vivo avec GS967 via administration orale à la dose de 1,5mg/kg/jour pendant 38 jours a significativement prolongé le taux de survie des rats Scn1a+/− et réduit la fréquence des crises[5]. Pendant deux jours consécutifs, l'administration quotidienne de 1.5mg/kg/jour de GS967 a significativement réduit la fréquence des crises chez les rats Scn2aQ54 et prévenu la perte des neurones du ganglion pulmonaire[6]. L'administration orale de 1mg/kg/jour de GS967 pendant 5 semaines consécutives a considérablement réduit la masse du ventricule gauche et l'oedème pulmonaire des rats Dahl sensibles au sel (DSS) soumis à un régime riche en sel, sans affecter la pression artérielle ou la contractilité du ventricule gauche[7].

Rats Scn1a+/−

Méthode de préparation

Le 18ème jour après la naissance (P18), les rats Scn1a+/− ont été sevés puis divisés aléatoirement en groupe GS967 ou en groupe témoin. Les rats du groupe traité avec GS967 ont reçu de la nourriture contenant GS967 (à la dose de 1,5mg/kg/jour). Le taux de survie des rats a été surveillé jusqu'à l'âge de 8 semaines. A 8 semaines, le traitement avec GS967 a été arrêté, et le taux de survie a continué d'être surveillé jusqu'à l'âge de 12 semaines.

Forme de dosage

1.5mg/kg/jour pendant 38 jours; p.o.

Domaines d'application

Le traitement avec GS967 a amélioré la survie des rats Scn1a+/−.
