Preparation Method

In order to determine the effect of oxaliplatin on cell viability, CAL27 cells and SCC25 cells were treated with different concentrations of oxaliplatin for 12, 24, and 48 hours.

Oxaliplatin inhibited cell viability, migration, and cloning formation of OSCC cells and induced cell death in vitro.

Preparation Method

Subjects were injected intraperitoneally (once every two days) with oxaliplatin (5 mg/kg in PBS, oxaliplatin group) or same volume of PBS (control group). After 21 days, mice were euthanized, and their xenografts were harvested and weighed.

5 mg/kg oxaliplatin for 21 days

Applications

Oxaliplatin inhibited tumor growth of OSCC and caused upregulation of PARP1 in vivo.

Background

Oxaliplatine est un médicament de chimiothérapie cytotoxique utilisé pour traiter le cancer. Oxaliplatine agit en perturbant le développement de l'ADN dans une cellule. Cela aide à traiter le cancer qui est causé par des cellules qui se développent rapidement et se divisent hors de contrôle.

Oxaliplatine peut induire des dommages à l'ADN dans les cellules cancéreuses. De plus, l'oxaliplatine peut interférer avec le cycle cellulaire, favoriser l'apoptose et induire l'autophagie dans les cellules tumorales[2,3]. L'oxaliplatine a inhibé la viabilité cellulaire, la migration et la formation de colonies de cellules OSCC et a induit la mort cellulaire in vitro[1]. L'oxaliplatine a inhibé la croissance des cellules HCCLM3 et Hep3B. La diminution des protéines anti-apoptotiques Bcl-2 et Bcl-xL ainsi que l'augmentation de la protéine pro-apoptotique Bax ont été observées lors de l'apoptose induite par oxaliplatine[6]. L'oxaliplatine s'est avérée active contre les lignées cellulaires C32 et G361 avec des valeurs IC50 de 49,48 et 9,07 uM (exposition d'une heure), 9,47 et 1,30 uM (exposition de quatre heures) ,et 0.98 and 0.14 uM (exposition pendant 24 heures), respectivement. À une exposition de 24 heures,l’oxalipatrine semble être significativement plus active que le cisplatine contre la ligne cellulaire G361 [4] . Entre 85% à88% du platine provenant d'Oxalipatrine était lié aux protéines plasmatiques au cours des cinq premières heures avec une demi-vie moyenne de1,71 +/- 0,06 h. Lorsque l'oxaliplatine a été incubée dans du sang entier, les érythrocytes ont absorbé 37,1 +/- 2,1% du platine total en deux heures (absorption maximale) qui n'était pas échangeable dans le plasma[5].

L'effet anticancéreux in vivo de l'oxaliplatine a été étudié chez une souris nue implantée sous-cutanément avec des cellules CAL27. Le volume moyen des tumeurs était nettement plus petit dans le groupe traité à l'oxaliplatine que dans le groupe témoin. Par conséquent, ces résultats ont indiqué que l'oxaliplatine inhibait la croissance des cellules xénogreffes du carcinome épidermoïde de la bouche et du pharynx in vivo, ce qui était accompagné d'une expression accrue de PARP1[1]. Les souris traitées à l'oxaliplatine ont présenté une prise de poids réduite, une allodynie mécanique et des déficits comportementaux exploratoires qui n'étaient pas significativement améliorés par l'exercice[7]. Lorsqu'on a étudié les toxicités hématologiques induites par l'oxaliplatine et la splénomégalie chez les souris, l'analyse sanguine a montré des diminutions dépendantes de la dose du nombre de globules blancs et rouges ainsi que des changements significatifs dans les indices hématologiques. Le poids de la rate était significativement augmenté, indiquant une splénomégalie, et le tissu pulmonaire rouge présentait une dysplasie substantielle[8]. Les souris traitées à l'oxaliplatine ont présenté une prise de poids réduite et des déficits comportementaux. Les souris traitées sur un cours plus court avaient une phosphorylation STAT3 significativement augmentée dans les muscles gastrocnémiens. Les souris recevant un traitement prolongé à l'oxaliplatine ont présenté une réduction de la masse musculaire des membres postérieurs avec une régulation à la hausse des gènes associés à la myopathie Foxo3, MAFbx et Bnip3[9].
