1-オレオイルリゾホスファチジン酸(LPA)は、様々な効果を持つ自然発生のリン脂質です。 LPAは多くの組織に広く分布しています。他の多くのバイオメディエーターと同様に、LPAはGタンパク質共役受容体など特定の細胞表面受容体を介して細胞と相互作用し、生物学的効果を引き起こします。
In vitro(試験管内):LPAはEdg/LPA受容体を介してシグナル伝達を仲介し、腫瘍細胞の増殖、移動、接着および抗アポトーシス機能を誘導する可能性があります。さらに、LPAはVEGFの上昇を引き起こすことが判明しており、悪性腫瘍の発生と進行に間接的な影響を与える可能性があります。 LPAはまたマトリックスメタロプロテアーゼ分泌や腫瘍血管新生因子の刺激も示唆されています[1]。
in vivo:LPAは、低酸素性肺高血圧症のラットモデルでその血管再構築効果を試験されました。低酸素群の動物から採取した血清は、ラット原発肺線維芽細胞に対するより高いケモアトラクタント特性を示し、この細胞移行増加はLPA受容体1および3拮抗剤によって防止されました。 LPAはまた、LPA受容体1または3の活性化を介して人間の肺動脈内皮細胞の接着特性も増加させることがわかりました[1]。
臨床試験:臨床結果により、LPA濃度は膵臓がん患者では有意に高くなることが示されました。膵臓がんの診断において、LPAの感度は89.6%であり、特異性は79.4%です。ただし、血漿中のLPA変化は、腫瘍サイズ、病理的段階、周囲リンパ節だけでなく特定の組織学的特徴とも有意な相関を示しています[2]。
