Description of 306-O12B

306-O12bは、脂質ナノ粒子(LNP)を生成するために使用されるイオン化可能なカチオン性資質である。肝臓固有のCRISPR-CAS9遺伝子編集に使える[1]。

306-O12B LNPはCas9 mRNAとSgangptl3を野生型C57BL/6鼠の肝細胞に特異的に送達し、38.5%の編集率の中央値と血清Angptl3タンパク質の対応する65.2%減少をもたらす。306-O12B LNPを使う送達はゴールドスタンダードLNPであるMC-3 LNPを使って、配達よりも効率だ[1]。

Protocol of 306-O12B

306-O12B LNP を使う体内ゲノム編集のプロトコル[1]:

1. 306-O12B LNPの合成:

ナノAssemblrマイクロ流体システムでLNPを調製される。

脂質、コレステロール、リン脂質(DOPC)、methoxypolyethylene glycol (DMG-PEG2000)は最終脂質濃度10 mg/mlで50/38.5/10/1.5のモル比で100%エタノールに溶解する。

次に脂質溶液をナノAssemblrマイクロ流体システムでmRNAを含んでいる酸性酢酸ナトリウム緩衝液(25 mM,pH 5.2)と混合する。306-O12b/mRNA重量比は7.5:1である

もらったLNPは4°Cで一晩、リン酸緩衝生理食塩水(PBS)(pH 7.4,10 mm)に対して透析する。

2.在体内基因组编辑中(例如:angptl3):

生体内の編集(例:angptl3):

6~8週齢のメスの野生型C57BL/6鼠にCas9 mRNAが1.0、2.0,2.0 mg/kgの用量で総mRNAが1.0、2.0 mg/kgの用量で静脈注射する。
