Description of 306-O12B

306-O12B is an ionizable cationic lipid used to generate lipid nanoparticles (LNPs). It can be used for liver-specific CRISPR-Cas9 gene editing[1].

306-O12B LNP specifically delivered Cas9 mRNA and sgAngptl3 to liver hepatocytes of wild-type C57BL/6 mice, resulting in a median editing rate of 38.5% and a corresponding 65.2% reduction of serum ANGPTL3 protein. Delivery using 306-O12B LNP was more efficient than delivery with MC-3 LNP, a gold standard LNP[1].

Protocol of 306-O12B

Protocol for in vivo genome editing using 306-O12B LNP [1]:

1.Synthesis of 306-O12B LNPs:

 LNPs were prepared using a NanoAssemblr microfluidic system.               

Lipidoids, cholesterol, phospholipids (DOPC), and methoxypolyethylene glycol (DMG-PEG2000) were dissolved in 100% ethanol at molar ratios of 50/38.5/10/1.5 at a final lipidoids concentration of 10 mg/mL.

The lipid solution was then mixed with an acidic sodium acetate buffer (25 mM, pH 5.2) containing mRNA by using the NanoAssemblr microfluidic system. The weight ratio of 306-O12B/mRNA was 7.5:1.

The resulting LNP was dialyzed against phosphate-buffered saline (PBS) (pH 7.4, 10 mM) overnight at 4 °C.

2.In vivo genome editing (eg:Angptl3):

Female wild-type C57BL/6 mice, aged 6 to 8 wk, were intravenously dosed with Cas9 mRNA and ANGPTL3-targeted sgRNA coloaded 306-O12B LNP at a dose of 1.0, 2.0, and 3.0 mg/kg in total RNA.
