アファチニブ（BIBW2992）は、不可逆的なErbB家族のチロシンキナーゼ阻害剤であり、表皮成長因子受容体（EGFR）/ヒト表皮成長因子受容体2（HER2）のキナーゼドメインにIC50値がそれぞれ0.5nMおよび14nMで結合することにより、ErbBシグナルをダウンレギュレートします。 ErbB受容体チロシンキナーゼファミリーは4つの細胞表面受容体から構成されており、ErbB1 / EGFR / HER1、ErbB2 / HER2、ErbB3 / HER3、およびErbB4 / HER4が含まれます。EGFRとHER2が特定の侵略性が高い癌や炎症関連疾患の発展と進行に重要な役割を果たすことが示されています。 アファチニブはA431、NIH-3T3-HER2、NCI-N87およびBT-474などのEGFR過剰発現およびHER2発現細胞株でEGF誘導性リン酸化及び細胞増殖を抑制することが示されています。 この成分はまた、EGFR / HER2の役割を研究するために様々な動物モデルで広く使用されています。アファチニブの経口投与は、キシェンオグラフトおよびトランスジェニック肺癌モデルでがん細胞の増殖と生存を抑制し、腫瘍退縮を抑制します。さらに、アファチニブはEGFR変異型非小細胞肺癌患者の治療に承認されたEGFRブロッカーとして特定されました。
