AG-1024は可逆的で、競合的で、選択的なインスリン様の成長因子-1受容体(IGF-1R)阻害剤であり、IC50値は7µMである[1]。AG-1024はインスリン受容体（IR）のリン酸化を57μMのIC50値で阻害できる[2]。AG-1024は細胞のアポトーシスを誘発でき、抗癌活性を持っている[3]。
体外では、獲得性のソラフェニブ耐性細胞株（SNU-sora-5とSK-sora-5細胞）をAG-1024 (5µM)で72時間処理した結果、ソラフェニブの細胞毒性を著しく増やし、細胞内mTOR/P21シグナル伝達経路を活性化し、細胞G1/S停止を促進した[4]。AG-1024 (10μM)でPC3とDU145細胞を処理した結果、細胞増殖を阻害し、細胞G2/M期の停止を誘発した[5]。
生体内では、AG-1024 (20mg/kg)を糖尿病腎障害モデルラットに8週間経口投与した結果、代謝障害を改善し、腎機能を改善し、腎炎症を減らし、腎線維症の形成を減らした[6]。

準備方法 | ラットをランダムに、対照、DN、DN+AG-1024とDN+メトホルミングループに分けた。各グループには15匹のラットがいる。一晩中断食させられたラットに、2%のペントバルビタールナトリウム（40mg/kg）を腹腔内注射し、右側の腎臓を除去した。対照群にクエン酸塩緩衝剤と0.5mLの生理食塩水を12週間毎日1回投与した。2週間後、DNグループのラットに、新鮮に調製されたストレプトゾシン（STZ；50mg/kg）を一回腹腔内注射した。4週間の培養の後、DNグループにある糖尿病ラットに毎日0.5mLの生理食塩水を投与し、DN+AG-1024グループのラットにAG-1024 (20mg/kg/日、経口)を投与し、DN+メトホルミングループに、メトホルミング(200mg/kg/日）を8週間経口投与した。 |
