Preparation Method |
Cells were stimulated on the second day of culture with 50 units/ml of recombinant mouse IL-2. After 7-9 d of culture, T cell blasts were used for in vitro activation and staining assays. Before stimulation, T cell blasts were incubated with 100 µM Cholesterol Sulfate or DMSO control in RPMI-1640 medium supplemented with 1% lipid-free BSA or 5% lipid-deficient FCS for 2 h at 37 ⊿ |

コレステロール硫酸塩は、1965年に初めて人間の血漿から単離され、100 mlあたり300µgの濃度で存在することが判明しました[1]。この最初の値の妥当性はすぐに確認され、限られた数の被験者を対象とした血漿中コレステロール硫酸塩レベルは174〜328 µg/100 mlに及ぶ報告がありました[2,3]。
コレステロール硫酸塩は、他のステロイド硫酸塩や非結合型コレステロールとは異なり、因子XIIを活性化することで固有の血液凝固系を引き起こす能力があります。 コレステロール硫酸塩は、因子XIIの存在下で前カリクレインを活性化します[4]。 コレステロール硫酸塩は、特にε、Η、およびζアイソフォームの多数の表皮タンパク質キナーゼCアイソフォームを活性化します[5]。 vitroでは、コレステロール硫酸塩はプラセボエステルよりも強力に蛋白質キナーゼCのΗアイソフォームを新規に活性化しました[6]。 血液凝固および線溶に重要な役割を果たすセリンプロテアーゼであるトロンビンおよびプラズミンは、コレステロール硫酸塩によって強力に阻害されます[7]。
コレステロール硫酸ナトリウム塩(100 µM 2 h)の前処理により、抗CD3誘導性のCD3ζリン酸化が著しく減少しました。コレステロール硫酸はTCRと特異的に相互作用して、シグナル伝達の下流成分を妨げることなく細胞膜内シグナル伝達を阻害します[8]。Cholesterol Sulfate処理された5C.C7 T細胞から抽出した(T細胞抗原受容体) TCRナノクラスターが強く減少しました[8]。マウスの胸腺内に20µl 25mMのコレステロール硫酸を注入することで、総胸腺細胞数が減少しました[8]。
