Description of DC-Chol (hydrochloride)

DC-Chol (hydrochloride)はAβ40線維の形成を抑制することができ、DC-Chol (hydrochloride)は酸化hCTのアミロイド形成を抑制することができる。

DC-Chol (hydrochloride)については、Aβ40(30 µM)と低濃度のDC-Chol (hydrochloride)(50 ng/mL)を共存させた像ではAβ線維が観察された。しかし、500 µg/mL DC-Chol (hydrochloride)存在下では、画像中に繊維構造は認められなかった。DC-Chol/DOPEカチオン性リポソームは、強いエントロピー駆動型の表面静電相互作用によって、CT-DNAを適切に凝縮し、コンパクト化する[3]。仔牛胸腺DNA(CT-DNA)とカチオン性脂質3β-[N-(N',N'-dimethylaminoethane)-carbamoyl]コレステロール塩酸塩(DC-Chol(hydrochloride))とDOPEからなる混合カチオン性リポソームから構成されるリポプレックスを解析した。DC-Chol/DOPEリポソームは、平均流体力学的直径が(120±10)nm程度であり、強いエントロピー駆動型の静電相互作用によって、カチオン性表面でDNA断片をコンパクトに凝縮する。50mol%のDODACまたはDC-Chol (hydrochloride)と20mol%のDSPE-PEG(2000)で調製した製剤は、6.5~12.5時間の循環半減期を示した。DSPE-PEG(2000)を用いて調製したDC-Chol (hydrochloride)製剤は、PEGを含まない製剤と比較して、s.c.HT29腫瘍に3倍蓄積した。DC-Chol (hydrochloride)は、マクロサイトーシスとクラスリンを介したエンドサイトーシスにより内在化される。

準備方法

Aβ40(30 μM)をpH 7.4のリン酸緩衝液に溶解し、Speci-Mixアリコットミキサーでインキュベートした。Aβ40を50 ng/mLのコレステロール-SO4、500 μg/mLのコレステロール-SO4、50 ng/mLのDC-Chol(塩酸塩)、および500 μg/mLのDC-cholesterolと共に処理した。

反応条件

50 ng/mLおよび500 μg/mLのDC-Chol(塩酸塩)をAβ40と共に17時間処理

アプリケーション

DC-Chol(塩酸塩)の場合、Aβ40(30 μM)と低濃度のDC-cholesterol(50 ng/mL)を共処理した画像でAβ線維が観察された。しかし、500 μg/mLのDC-cholesterolが存在する場合、線維構造は画像内に見られなかった。
