Polybreneは第四紀アモンニウム塩であり、ヘパリン中立剤として最初に臨床診療に導入される。なお、低用量のPolybreneは体内と体外ではウイルス媒介遺伝子導入の効率的を促進するために一般的に使用される[2-5]。Polybreneは反対側の二重層間の静電反発を中和することにより、細胞表面上のウイルスの結合を促進できる[6-7]。
Polybrene(0-32 µg/ml; 24H)は濃度依存の効果を示している。4 µg/mlの低い濃度から始まると明確な神経炎のビーズが出現するが、8 µg/mLの濃度は神経炎の断片化を誘発する[8]。Polyethylene(8µg/mL)は間葉系幹細胞(MSC)の増殖速度を有意に低下させる[9]。Polybreneは脂肪分解の効率を著しく向上させ、さんざまな細胞株における血清媒介脂肪症の阻害を補償する[10]。
Polybrene(4 µg/µl iCV)は注射は3日目に運動機能阻害を示す鼠を引き起こす[8]。
