Description of Binuclease

Binuclease is a non-specific nuclease derived from Serratia marcescens (also known as Bacillus thuringiensis) modified by protein engineering technology. Binuclease does not contain bacterial endotoxins expressed by prokaryotes.

Binuclease unit activity definition: One unit of enzyme activity is the amount of oligonucleotide produced within 30 minutes under the reaction conditions of 37ºC and pH 8.0, which is equivalent to the amount of oligonucleotide that changes the 260nm absorbance by 1.0. This product is a lyophilized powder with an enzyme activity value of ≥20kU/mg.

Functionally, Binuclease can cleave various forms of DNA and RNA (including single-stranded, double-stranded, linear, circular or supercoiled forms of DNA and RNA), and can cut phosphodiester bonds to produce 5'-phosphate nucleotides or 5'-phosphate oligonucleotides. Binuclease has no specific requirements for nucleic acid base sequences and can cut between any nucleotides in the chain. It can be used to remove nucleic acids in biological products. In the vaccine industry, protein and polysaccharide pharmaceutical industry, it is widely used to remove host nucleic acid residues, which can reduce the nucleic acid contamination of vaccines and protein products to the pg level. Binuclease can reduce the viscosity of cell lysate by degrading nucleic acids, which helps promote solution filtration/ultrafiltration during the purification of cell-derived particles such as viruses, AAV vectors and inclusion bodies, shorten processing time, improve the separation efficiency of precipitates and supernatants during centrifugation, and improve chromatographic purification efficiency, ultimately achieving the effect of increasing yield and effectively improving the purity of the final product. In bioanalysis applications, it can be used in ELISA sample preparation, chromatography, biphasic electrophoresis (protein mapping) and blotting analysis to improve resolution and sample recovery.

During the use of Binuclease, you need to prepare the dilution buffer and storage buffer by yourself. The specific configuration scheme is as follows:

Dilution buffer: 20mM Tris-HCl (pH 8.0), 2mM MgCl2 , 20mM NaCl

Storage buffer: 20mM Tris-HCl (pH 8.0), 2mM MgCl2 , 20mM NaCl, 50% glycerin

Binuclease application advantages:

Binuclease can maintain high stability and activity under various extreme industrial conditions, can reduce unit costs through large-scale mass production, and can perfectly replace expensive Benzonase and other similar products for research and production in the biopharmaceutical industry.

The following commonly used reagents have an inhibitory effect on the activity of broad-spectrum nucleases. Within the following concentration range, the loss of broad-spectrum nuclease activity is within 50%. Please use with caution.。
