2' ,3' -cGAMP는 STING 수용체의 특정 작용제이고 그 작용 메커니즘은 주로 STING 신호 경로를 활성화하여 인터페론 및 염증 사이토카인의 생성을 촉진하는 것이다. 2',3'-cGAMP는 포유류의 선천 면역 경로에서 중요한 역할을 하고 세포질 DNA를 감시하는 역할을 한다. 2',3'-cGAMP가 STING에 결합하는Kd 값은 3.79 nM이다[1]. 2',3'-cGAMP는 면역 조절, 항바이러스 면역 및 암 치료 연구에 일반적으로 사용된다.
체외 실험에서 2',3'-cGAMP(2.2, 4.4, 8.75, 17, 35 μM)가 PC-3 세포에 48시간 처리된 후의 IC50 값은 11.46 μM이다[2]. 2',3'-cGAMP(4 µg/ml)로 PC-3 세포 라인을 10분간 처리하면 STING을 활성화한다[3].
체내 실험에서 2',3'-cGAMP의 10 μg가 C57 BL/6 쥐의 멜라놈마를 싸우는데 200 μg의 PD-L1 항체만큼 효과적이다. 적정 실험은 2',3'-cGAMP가 PD-L1 항체의 항종 효과를 용량 의존적인 방식으로 증강시킨다고 보여준다[4].
