4-Methylumbelliferyl-β-D-Glucopyranoside is a β-D-glucoside that serves as a fluorescent substrate for β-glucosidase, thereby allowing for the detection and analysis of this enzyme's activity[1]. The enzymatic cleavage of 4-Methylumbelliferyl β-D-glucopyranoside results in the release of the highly fluorescent compound 4-Methylumbelliferone. The maximum emission wavelength of 4-MU ranges from 445 to 454nm, while its excitation wavelength varies with pH: at pH 4.6, 7.4, and 10.4, the corresponding excitation wavelengths are approximately 330nm, 370nm, and 385nm, respectively[2].

Assay of Proteasome Activity in Cell

Enzyme Sample Preparation: Lyse cells and collect the supernatant. Store the clarified lysate at 4°C for subsequent use.
β-Glucosidase Activity Detection: Prepare a reaction mixture containing 5μM 4-Methylumbelliferyl-β-D-Glucopyranoside, 100mM citrate buffer (pH=5.0), total volume: 200μL; Incubate at 45°C for 1 hour. Terminate the reaction by adding 2.5mL of 0.5M Na₂CO₃ buffer (pH=10.4). Detect the product using a fluorometer with an excitation wavelength of 350nm and an emission wavelength of 440nm.

1) Free fluorescent substrates are subject to quenching. Please try to avoid light exposure to slow down fluorescence quenching.
2) For your safety and health, please wear a lab coat and disposable gloves when operating.
