Description of BODIPY-aminoacetaldehyde diethyl acetal

BODIPY-aminoacetaldehyde diethyl acetal (BAAA-DA)은 BODIPY-aminoacetaldehyde (BAAA)의 안정된 전이물로 세포를 관통할 수 있는 형광 전구체로서 알데하이드 드로게나이즈 (ALDH)를 탐지하는 데 사용되고 투과성 형광 파장이 488/512nm이다.

BODIPY-aminoacetaldehyde diethyl acetal은 산성 조건에서 BAAA로 변환된다. BAAA는 전하를 띠지 않고 세포 투과성으로 온전한 살아있는 세포의 플라즈마막을 자유롭게 관통할 수 있는 세포 투과성을 가지고 있다. BAAA는 세포 내에서 ALDH에 의해 Bodipy-아미노 이산수산 (BAA)로 변환되고 BAA의 순 음전하로 인해 세포에 의하여 보유되어ALDH 활동이 높은 세포를 식별하는 데 사용될 수 있다[1-2]. BODIPY-aminoacetaldehyde diethyl acetal은 고 ALDH 활동을 가진 인간 혈액 포세슬 기원 세포를 분리하는 데 사용되었고 흐름 사이토메트리로 고 수준의 ALDH를 포함하는 암 기원 세포를 정리할 수도 있다[1, 3]. BODIPY-aminoacetaldehyde diethyl acetal은 살아있는 ALDHpos 세포를 특성화하는 데 사용될 수 있고 세포에 사용된 BAA는 488nm에서 흥분하여 512nm의 최대 발산 파장으로 사용될 수 있다[4].

Protocol of BODIPY-aminoacetaldehyde diethyl acetal

이 실험은 문헌에서 나왔고 구체적인 필요에 따라 수정하주시기 바랍니다.

1. BODIPY-aminoacetaldehyde diethyl acetal 염색 용액 준비.

(1) BODIPY-aminoacetaldehyde diethyl acetal염료 저장 용액 준비: 고체 분말을 DMSO로 녹여 BODIPY-aminoacetaldehyde diethyl acetal 염료 저장 용액을 준비합니다.

참고: BODIPY-aminoacetaldehyde diethyl acetal저장 용액을 별도 분쇄 후 -4℃나 -20℃의 어두운 곳에 보관하여 반복 동결과 해동을 피하도록 저장합니다.

(2) 작업 용액 준비: BODIPY-aminoacetaldehyde diethyl acetal을 BAAA로 변환하기 위해 동등한 용량의 2N HCl을 추가합니다. 2시간 후 적당한 버퍼 (예: PBS)를 추가하여 농도가 1-20μM인 BODIPY-aminoacetaldehyde diethyl acetal작업 용액을 준비합니다.

참고: 실제 조건에 따라 BODIPY-aminoacetaldehyde diethyl acetal 작업 용액의 농도를 조정하고 사용하기 전에 준비해 주십시오.

2.세포 염색

수정세포: 수정세포를 1000g로 3-5분간 원심분리하고 상층액을 버리고 PBS로 두 번 5분씩 세척합니다.

접착세포: 트립신을 추가하여 세포를 분해한 후 1000g로 3-5분간 원심분리하고 상층액을 버리고 PBS로 두 번5분씩 세척합니다.

(1) 수집된 세포에 BODIPY-aminoacetaldehyde diethyl acetal 작업 용액 1mL을 추가하고 37℃ 암광에서 30-60분간 포화시킵니다.

(2) 포화 후 1000g로 5분간 원심분리하고 상층액을 제거한 후 PBS로 2-3번5분씩 세척합니다.

(3) 세포를 PBS로 재수정하고 흐름 사이토메트리로 분석합니다 (Ex/Em은 488/512nm입니다).

참고:

① 이 제품은 비활성화된 형광 염료로서 먼저 산성용액(2N HCl)으로 활성화해야 합니다.

② 모든 형광 염료에는 퀴촨 현상이 있으므로 빛을 피해주세요.

③ 안전과 건강을 위해 수술시 실험가운과 일회용 장갑을 착용해 주십시오.
