Applications

Polybrene displays concentration-dependent effects. Starting at concentrations as low as 4 µg/mL, distinct neuritic beads emerge, whereas a concentration of 8 µg/mL induces neuritic fragmentation.

Preparation Method

4 µg/µL polybrene was incubated at 37℃ for 15 min, and intracerebroventricularly (ICV) injection of 5 µL mixture was performed for 20 min. Rotating Rod test will take place in three days.

Applications

Polybrene injection resulted in mice exhibiting motor dysfunction on the third day.

Background

폴리브렌은 사분자 암모늄 염으로, 먼저 헤파린 중화제로 임상 실습에 도입되었습니다[1]. 또한 낮은 용량의 폴리브렌은 생체 내외에서 바이러스 매개 유전자 전달의 효율성을 증진시키는 데 널리 사용되었습니다[2-5]. 폴리브렌은 상호 대립하는 이중층 사이의 정전기적 척력을 중화하여 바이러스가 세포 표면에 결합하는 것을 촉진합니다[6-7].

Polybrene(0-32 µg/mL;24h)는 농도에 따라 다른 효과를 나타냅니다. 4 µg/mL부터 뚜렷한 신경 섬유 구슬이 나타나며, 8 µg/mL의 농도에서는 신경 섬유 파편화가 유발됩니다[8]. Polyethylene(8µg/mL)은 중간엽 줄기세포(MSCs)의 증식 속도를 현저하게 감소시켰습니다[9]. Polybrene은 다양한 세포주에서 리포펙션(lipofection)의 효율을 크게 향상시키고, 리포펙션에 대한 혈청 매개 억제 작용을 보완합니다[10].

폴리브렌(4 µg/µL; ICV) 주사는 쥐들이 3일째 운동 기능 장애를 나타내게 했습니다. 폴리브렌은 쥐에서 신경 세포의 사멸과 젤라이트 형성을 유발합니다[8].
