Preparation Method

In order to determine the effect of oxaliplatin on cell viability, CAL27 cells and SCC25 cells were treated with different concentrations of oxaliplatin for 12, 24, and 48 hours.

Reaction Conditions

0-300uM Oxaliplatin for 12, 24, and 48 hours

Applications

Oxaliplatin inhibited cell viability, migration, and cloning formation of OSCC cells and induced cell death in vitro.

Preparation Method

Subjects were injected intraperitoneally (once every two days) with oxaliplatin (5 mg/kg in PBS, oxaliplatin group) or same volume of PBS (control group). After 21 days, mice were euthanized, and their xenografts were harvested and weighed.

5 mg/kg oxaliplatin for 21 days

Applications

Oxaliplatin inhibited tumor growth of OSCC and caused upregulation of PARP1 in vivo.

Background

옥살리플라틴은 암을 치료하기 위해 사용되는 세포독성 항암제입니다. 옥살리플라틴은 세포 내의 DNA 발달에 방해를 주어, 무절제하게 빠르게 자라고 분열하는 암세포를 치료하는데 도움이 됩니다.

옥살리플라틴은 암세포에서 DNA 손상을 유발할 수 있습니다. 또한, 세포주기에 간섭하고 종양세포에서 자가소멸을 촉진하며, 자가식을 유도할 수 있습니다[2,3]. 옥살리플라틴은 OSCC 세포의 세포 생존율, 이동성 및 클론 형성 억제를 일으켰으며 vitro 상에서 세포 사망을 유도시켰습니다[1]. 옥살리플라틴은 HCCLM3 및 Hep3B 세포의 성장을 억제합니다. Bcl-2와 Bcl-xL 항아척 단백질의 다운레귤레이션과 Bax 프로아파치닉 단백질의 업레귤레이션이 옥살리 플라 틴으로 인한 아파 토 시스 도중 관찰되었습니다[6]. C32 및 G361 세 평면에 대해 1 시간 노출시 IC50 값이 각각 49.48 밀몰 / L과 9.07 uM이고, 4 시간 노출시 각각 9.47 밀몰 / L과 1.30 uM이며, 24 시간 노출시 각각 0.98 밀몰 / L과 0.14 uM입니다. G361 세포주에 대해 옥살리 플라 틴은 시스플라틴보다 유의하게 활성화되는 것으로 나타났습니다[4]. 옥살리 플라 틴에서 모든 발레늄 중 85-88 %가 최초의 5 시간 이내에 혈장 단백질에 결합되었습니다. 평균 반감기는 1.71 +/- 0.06 h였습니다. 전혈 내에서 옥살리플라틴을 배양 할 때, 적혈구는 총 발레늄의 약37.1 +/-2 .1%를 최대 흡수하는 것으로 나타났으며, 이것은 혈장으로 교환될 수 없었습니다[5].

옥살리플라틴의 생체 내 항암 효과는 CAL27 세포가 주입된 누드 마우스를 이용하여 연구되었다. 옥살리플라틴 군에서 종양의 평균 부피는 대조군에 비해 현저하게 작았다. 따라서 이러한 결과는 옥살리플라틴이 생체 내 외래 유래 구강 경부 암세포의 성장을 억제하는 것을 나타내며, PARP1 발현 상승과 동반되었다[1]. 운동으로 크게 개선되지 않은 증상인 체중증가 감소, 기계적 전도성 통증 및 탐사 행동 결함이 옥살리플라틴 처리된 마우스에서 관찰되었다[7]. 마우스에서 옥살리프란 유도된 혈액학적 독성 및 비대성 조사 시, 혈액 분석 결과 백혈구와 적혈구 수가 용량 의존적으로 감소하고, 혈액학 지수에 중요한 변화가 있음을 보여주었다. 비대성은 중요하게 증가하였으며, 붉은색 조직에서는 심한 이형성이 나타났다[8]. 옥살리플라틴 처리된 마우스는 체중증가 감소 및 행동 결함을 보였다. 짧은 기간 동안 치료받은 마우스들은 gastrocnemius 근육에서 STAT3 인산화가 유의하게 증가하였다. 연장된 옥살리플라틴 치료를 받은 마우스들은 Foxo3, MAFbx 및 Bnip3와 같은 근병증 관련 유전자의 상승과 함께 후지 다리 근육량이 감소되었다[9].
