Description of CAY10732

CAY10732 funciona como un agente fluorescente diseñado para identificar la presencia de monóxido de carbono.1 Cuando se expone al paladio (Pd) y al monóxido de carbono, CAY10732 inicia la reacción de Tsuji-Trost mediada por Pd0, lo que resulta en la liberación de 2,7-dichlorofluoresceína. La fluorescencia emitida por este compuesto sirve como indicador de la producción de monóxido de carbono, tanto en entornos de laboratorio controlados como dentro de células vivas. Los picos de excitación/emisión de la 2,7-dichlorofluoresceína se miden a 493/527 nm, respectivamente[1].

Protocol of CAY10732

Protocolo para imágenes de CO ien células vivas[1]

Preparación de soluciones de stock:Las soluciones de stock de CAY10732 (2 mM), PdCl2 (2 mM), y CORM-3 (10 mM) se prepararon por separado en DMSO de grado HPLC y se utilizaron frescas. SE preparó CAY10732 (1 μM) y PdCl2 (1 μM) en tampón de PBS (10 mM, pH 7.4, con 0.5% de DMSO, v/v) en use.

1.Las células de HeLa se sembraron en una placa de cultivo de 24 pozos durante una noche antes de los experimentos de imagen celular.

2.En un experimento típico de imágenes celulares, como controles, las células vivas se incubaron con CAY10732 (1 μM) y una mezcla de CAY10732 y PdCl2 (1 μM cada uno) a 37℃ durante 30 minutos, respectivamente, y se les fotografió después de lavar con PBS tres veces.

3.Para las imágenes de CO exógeno, las células HeLa se pretrataron con CORM-3 (1, 5, y 10 μM, respectivamente) durante 30 minutos a 37℃ y luego se incubaron con una mezcla de CAY10732 y PdCl2 (1 μM cada una) durante 30 minutos a 37℃. Después de lavar las células con tampón PBS, se tomaron imágenes de las células.

4.Para las imágenes de la estimulación del hemo producida por CO, las células HeLa se pretrataron con hemo (100 μM) durante 30 minutos a 37℃ y luego se incubaron con una mezcla de CAY10732 y PdCl2 (1 μM each) durante 30 minutos a 37℃. Después de lavar las células con tampón PBS, se tomaron imágenes de las células.
