Description of GMax™ in vivo RNA Transfection Reagent (Local Effect)

GMax ™  in vivo RNA Transfection Reagent (Local Effect) is a novel ready to use lipid nanoparticle (LNP) product suitable for in vivo transfection of mRNA to experimental animals. Lipid Nanoparticles (LNP) is a nanoscale drug delivery system based on lipids, widely used for the delivery of nucleic acids (such as mRNA, siRNA, DNA, etc.) and small molecule drugs. This kit utilizes the unique transmembrane transport and escape mechanism of LNP to achieve efficient delivery of RNA, suitable for in vivo transfection of mRNA and short stranded RNA to experimental animals, including but not limited to intramuscular injection, intravenous injection, subcutaneous injection, intraperitoneal injection and other administration routes. This kit is suitable for Local Effect types of RNA functions.

Protocol of GMax™ in vivo RNA Transfection Reagent (Local Effect)

1. In vivo transfection procedure (taking a 20g mouse and subcutaneous injection of 20μg mRNA as an example)

1) Preparation of mRNA premix: Take 20μg of mRNA (original concentration: 1μg/μL) and place it in an RNase free Ep tube. Add 160μL buffer for dilution and mix well.

2) Preparation of mRNA LNP complex (mRNA concentration 0.1μg/μL): Add20 μL of LNP nanosuspension to the above mRNA premix, gently mix with a pipette at least 10 times, and mix evenly.

Note: The mRNA LNP complex can be stored stably at room temperature for at least 2 hours.

3) Subcutaneous injection: Inject 200μL of the mRNA-LNP complex subcutaneously into mice.

2. Recommended dosage for different administration routes

1) The dosage of products for different administration routes can be prepared according to Table 1.

2) Conversion of dosage for each component added

a. The actual amount of mRNA delivered should be evaluated comprehensively based on animal models, administration routes, and targeted organs.

b. The recommended dosage ratio of mRNA to LNP nanosuspension is 1:1 (μg mRNA: μL LNP).

c. Buffer dosage: VBuffer=Vinjection dose - VmRNA -VLNP, and the mRNA concentration in the mRNA LNP complex should not exceed 0.2μg/μL

3. Precautions

1) Please perform complex preparation operations in a clean environment and ensure that the consumables used during the operation are sterile and enzyme free.

2) Please ensure that the solvent in the mRNA stock solution is RNase free water, and the concentration of mRNA is not less than 1 μg/μL, in order to achieve better transfection efficiency; For homemade mRNA, additional washing is required to remove residual salt and prevent precipitation.

3) The positive control for Fluc mRNA is freeze-dried powder, please centrifuge before opening the lid. Dissolve in enzyme free sterile water to prepare a positive control solution of 1 μg/μL. The solution can be stored at -80 ℃ for 6 months to avoid repeated freezing and thawing.

4) Do not place the mRNA LNP complex under freezing conditions.

5) The product is for scientific research use only.
