El esfingosina 1-fosfato es un producto metabólico de los esfingolípidos de la membrana celular. Está unido a chaperonas extracelulares, enriquecido en fluidos circulatorios y se une a receptores acoplados a proteínas G del esfingosina 1-fosfato (Sphingosine 1-phosphateRs) utilizados para regular el desarrollo embrionario, la función de órganos postnatales y enfermedades.[1]
In vitro, el tratamiento con 200 y 400 μM de esfingosina-1-fosfato en muestras corticales ováricas humanas disminuyó de manera dependiente de la dosis la expresión proteica de caspasa-3 clivada mediante western blot y el número de folículos apoptóticos teñidos positivamente para caspasa-3 clivada mediante inmunohistoquímica. Además, in vitro, con una concentración dependiente del Sphingosine 1-phosphate entre 0.1-10 μM se evocó la generación potencial (AP) por grabaciones electrofisiológicas de fibra única. Además, utilizando experimentos de imagen Ca2+, Sphingosine 1-phosphate a 1 μM puede provocar un aumento transitorio en el Ca2+ intracelular en los astrocitos seguido por una elevación sostenida.
En vivo, la superfusión de 0.001-10 μM de esfingosina 1-fosfato provocó una vasoconstricción dependiente de la concentración en microvasos preglomerulares, predominantemente arteriolas aferentes. En un experimento in vivo se demostró que la inyección intravenosa de 10 mg/kg de esfingosina 1-fosfato en ratones causó rigidez e muerte inmediata. En beagles machos, el tratamiento con 85 ug/kg de esfingosina 1-fosfato por vía intravenosa disminuyó la formación del cociente Q(s)/Q(t) (32%), y tanto la presencia de proteínas (72%) como los neutrófilos (95%) en el líquido BAL en comparación con los controles tratados con vehículo. Sin embargo, la esfingosina 1-fostato aumentó la producción sistémica inducida por LPS de tres citocinas inflamatorias: TNF-alfa (6 veces), KC (1,2 veces) e IL-6 (3 veces), sin resultar en disfunción endorgánica.
