Mesure de l'activité estérolytique du facteur D du complément à l'aide de Z-Lys-SBzl . HCl[1]

1. Préparation des réactifs: Dissoudre Z-Lys-SBzl . HCl dans le tampon de réaction pour une utilisation ultérieure. Préparer un tampon Hepes 0.10M 0.50M de NaCl et 9.8% de DMSO, et une solution de 4,4'-dithiodipyridine.
2. Configuration du système réactionnel: Dans une cuve, établir un système réactionnel final avec un volume de 1mL. Le système contient le tampon Hepes à 0.19mM, et 1μM de facteur D humain purifié du complément .
3. Détection de l'activité enzymatique: Placer la cuve dans un spectrophotomètre et surveiller le changement d'obsorbance à 324nm à 27°C. Le benzyl mercaptan libéré par l'hydrolise de Z-Lys-SBzl·HCl par le facteur D réagit immédiatement avec la 4,4’-dithiodipyridine. Le coefficient d'extinction molaire (ε) de ce produit à 324nm est de 19,800M⁻¹ cm⁻¹. La vitesse initiale de la réaction enzymatique (v₀) est calculée à partir de la vitesse initial de l'augmentation de l'absorbance.
Précautions:
(1) Pour éviter l'achèvement de la réaction, une quantité excédentaire de trypsine (ex., 5μg) peut être ajoutée au système après la mesure de al vitesse initiale pour hydrolyser complètement le substrat restant en un court laps de tamps, ce qui permet un calcul précis de la concentration initiale du substrat. (2) Pour votre sécurité et votre santé, veuillez porter une blouse de laboratoire et des gants jetables pendant l'opération.
