Il est largement utilisé dans la synthèse d'intermédiaires pharmaceutiques et de monomères de matériaux[1]. Du fait de sa forte réactivité, il doit être conservé à basse température et à l'abri de la lumière, et des mesures de protection doivent être prises pendant l'opération.

Ce protocole utilise l'expérience de synthèse de l'acide 2-lévinique à l'aide de Ethyl 2-acetoxy-2-methylacetoacetate[1-2] comme exemple. Il ne sert qu'à titre de guide expérimental. L'application réelle doit être modifiée en fonction de vos besoins.
1.Préparation des réactifs : Ethyl 2-acetoxy-2-methylacetoacetate; acide sulfurique (H₂SO₄, 10%); bicarbonate de sodium (NaHCO₃, solution saturée); sulfate de magnésium anhydre (MgSO₄, désiccant); éther (solvant d'extraction).
2.Hydrolyse acide : Ajouter Ethyl 2-acetoxy-2-methylacetoacetate (5.0g, environ 24mmol) et 10% H₂SO₄ (30mL) dans un ballon à fond rond par ordre et secouer pour mélanger. Connecter le ballon à fond rond à un réfrigérant à reflux, chauffer à 80-90°C et arrêter le chauffage après avoir agité pendant 4-6 heures. Le mélange réactionnel a été refroidi à la température ambiante, le produit a été extrait avec de l'éther (3×20mL) et les phases organiques ont été combinées. La phase organique a été lavée avec une solution saturée de NaHCO₃ jusqu'à neutralité. Après séchage avec MgSO₄ anhydre, le solvant a été éliminé par évaporation rotative après filtration pour obtenir un produit brut.
3.Réaction de décarboxylation : Le produit brut a été dissous dans une petite quantité d'eau, chauffé à 100°C pendant 1 - 2 heures pour promouvoir la libération de CO₂. Après refroidissement, il a été à nouveau extrait avec de l'éther, séché et concentré pour obtenir de l'acide 2-lévinique.
4.Purification et stokage : L'acide 2-lévinique a été distillé sous pression réduite, et la fraction ayant un point d'ébullition de 120-130°C/10mmHg a été collectée, dissoute dans de l'éthanol chaud, refroidie et cristallisée, puis filtrée et séchée pour obtenir le produit pur. Le produit pur a été scellé et stocké à 4°C (court terme) ou à -20°C (long terme) de manière à l'abri de la lumière pour éviter l'absorption d'humidité.
Précautions:
1.L'équipement de protection (gants, lunettes de protection) doit être porté lors des opérations dans les conditions fortement acides.
2.Chauffer lentement pendant l'étape de décarboxylation pour éviter un ébullition violente.
3.Pour votre sécurité et votre santé, portez un tablier de laboratoire et des gants jetables lors des opérations.
4.Eviter d'inhaler les vapeurs et opérer dans une hotte à fumée.
