Abz-Val-Ala-Asp-Nva-Arg-Asp-Arg-Gln-EDDnp (trifluoroacetate salt)は蛍光共鳴エネルギー移動(FRET)基を含有する修飾ペプチドである。320nmの励起波長と420nmの発光波長で蛍光シグナルをモニタリングでき、Abz-Val-Ala-Asp-Nva-Arg-Asp-Arg-Gln-EDDnpは、プロテアーゼ活性を検出する蛍光基質としてよく使われる[1]。

細胞におけるプロテアソーム活性の測定
このプロトコールは研究データから整理されたものであり、参考までです。特定の実験需要に基づいて調整が必要な場合があります。
(1)試薬の調製:Abz-Val-Ala-Asp-Nva-Arg-Asp-Arg-Gln-EDDnp凍結乾燥粉末をジメチルホルムアミド(DMF)で溶解し、高濃度の貯蔵液(例えば5mM)を調製し、アリコートして、-20°Cで保存する。
(2)酵素サンプルの調製:精製のPR3酵素(濃度範囲は通常0.1nMから10nM)またはPR3を含有する細胞懸濁液(例えば活性化された好中球)を使う。
(3)PR3酵素活性の検出:反応混合物には20μMのAbz-Val-Ala-Asp-Nva-Arg-Asp-Arg-Gln-EDDnp、50mMのHEPES、150mMのNaClと0.05%のTriton X-100が含まれており、総体積は200μLである。37℃で30分間インキュベートする。マイクロプレート分光蛍光光度計で蛍光をモニタリングする。Abzに対する励起波長は320nmであり、発光波長は420nmである。
注意事項:
(1)遊離の蛍光基質には消光問題がある。蛍光消光を遅らせるためにはできる限り光への暴露を避けて下さい。
(2)DMFは酵素の活性を影響するかもしれない。貯蔵液におけるDMFの最終濃度をできるだけ低い水準に維持して下さい(通常は<1%)。
(3)ご自分の安全と健康の為に、実験の間には白衣と使い捨ての手袋をご着用下さい。
