SNAP es donante de óxido nítrico (NO), empleado como herramienta farmacológica para estudiar las funciones fisiológicas y farmacológicas del óxido nítrico[1]. SNAP se descompone de forma espontánea o bajo la influencia de la luz, el calor o iones metálicos, liberando óxido nítrico y generando el disulfuro correspondiente [2]. Este compuesto permite estudiar los mecanismos de regulación cardiovascular, la neurotransimisión y la respuesta inmune[3-4].
In vitro, el tratamiento con SNAP (1mM) durante 8 horas indujo de froma citotoxicidad en células de hepatoma de rata Fu5 [5]. La exposición de células de carcinoma pulmonar de Lewis (LLC-F6) y células de melanoma B16 (B16-4A5-F6) a SNAP (2mM) durante 24 horas redujo la viabilidad de ambos tipos celulares[6].
In vivo, una única inyección intramuscular de SNAP (1.0mg/kg) administrada 3 horas después de inducir la isquemia crítica de la extremidad (CLI)en las SD aumentó de forma la relación de flujo sanguíneo entre la extremidad isquémica y la normal (INBF) el día 14, incluyendo la densidad microvascular en la zona isquémica[7]. La infusión continua intracerebroventricular de SNAP (0.1μM; 2μL) durante 6 días en ratones P2X7R+/+ and P2X7R−/− elevó de manera la S-nitrosylation de la proteína disulfuro isomerasa (SNO-PDI) en el hipocampo aumentó y la fosforilación de NF-κB p65 en la serina 276 en la microglía[8].
