Clesitamig (ALPS-12) is a trispecific T cell engager with two CD3 / CD137 dual-specific Fabs and one DLL3 Fab. Clesitamig engages CD3 and CD137 on T cells to activate and co-stimulate T cells, promoting their proliferation, survival, and antitumor activity. Clesitamig binds to DLL3 on tumor cells for tumor-specific recognition. Clesitamig can be used for the research of small cell lung cancer (SCLC) [1] [2].
In Vivo, Clesitamig inhibits tumor growth in mouse SCLC xenograft models[2].
In Vitro, Clesitamig successfully engages its targets to activate CD3 and CD137 signaling, as demonstrated by a dose-dependent increase in NFAT and NF-κB activity in Jurkat reporter cells cocultured with DLL3-positive cells[2]. Clesitamig induces cytotoxicity against SCLC cell lines co-cultured with human PBMCs, with EC50 values in the two-digit picomolar range[2].
