Z223-2027 is a FcRn inhibitor. Z223-2027 competitively blocks the binding of immunoglobulin G ( IgG ) to FcRn. Z223-2027 can be used in the research of IgG-mediated autoimmune diseases [1].
In Vitro, Z223-2027 acts as an FcRn antagonist, exerting its function by competitively blocking the binding of IgG to immobilized human FcRn[1]. Z223-2027 stably binds to human FcRn (PDB: 5WHK) with a binding free energy of −27.8 kcal/mol, and interacts with the key residues D130 and W127, which supports its mechanism as an FcRn antagonist by blocking IgG binding[1].
