Description

Acetocarmine (Aceto Carmine) is a commonly used alkaline dye used for nuclear staining, chromosome fixation, and staining. To observe the mitosis of plant cells, chromosomes need to be stained with alkaline solution, and acetic acid magenta or gentian violet staining solution can be chosen. To observe whether the pollen is in the mononuclear stage, acetic acid carmine can be used for pollen detection.
Acetic acid carmine solution, also known as acetic acid carmine staining solution or acetic acid carmine staining solution, is mainly composed of high concentrations of acetic acid, carmine, etc., with an acidic pH value. Mainly used for staining the nucleus of samples such as pollen, animal and plant tissue slices, and can also be used for mitochondrial staining.

Matters needing attention

1) This product needs to be tightly sealed and stored after each use, otherwise the dyeing efficiency will be reduced.

2) This product contains weak acids and a pungent odor. Please take protective measures.

3) For your safety and health, please wear laboratory clothes and disposable gloves when operating.

Operation steps (for reference only)

1. For pollen particles
Take a small amount of pollen and place it on a glass slide. Add an appropriate amount of acetic acid magenta staining solution and mix thoroughly. Immediately cover the slide and stain for 10-30 minutes. Remove excess liquid and observe under a microscope immediately.

2. For animal and plant slicing
Routine processing of slices (such as dewaxing to water), adding an appropriate amount of acetic acid magenta staining solution to stain for 5-20 minutes, rinsing with tap water, sealing, and immediately observing under a microscope.

3. For some fresh plant tissues (such as onion epidermis)
Directly immerse in acetic acid magenta staining solution for 5-20 minutes, rinse with tap water for 2-3 minutes, place on a slide and cover with a cover glass, and immediately observe under a microscope.

[Note]: If the staining effect is not good, please quickly bake back and forth a few times on an alcohol lamp, in order to destroy the chromatin and make the nucleus stain.
