Inosinic acid (IMP)est un nuléotide qui joue un rôle central dans le métabolisme des purines et qui constitue également un exhausteur de goût important dans l'induistrie alimentaire[1]. Inosinic acid peut être converti en adénosine monophosphate (AMP) et en guanosine monophosphate (GMP), et il participe aux processus de métabolisme énergétique cellulaire[2]. Utilisé comme additif alimentaire (E630), Inosinic acid potentialise significativement la saveur umami des aliments lorsqu'il est associé au glutamate monosodique, et trouve des applications étendues dans les additifs pour aliments du bétail[3]. Inosinic acid présente également un intérêt scientifique croissant dans les domaines de la régulation immunitaire et de la neuroprotection[4].
In vitro, le traitement des cellules de moelle osseuse lineage⁻/low avec Inosinic acid (15mM) pendant 15–30 minutes active les voie de signalisation pAkt et pS6, stimulant la régulation positive de l'expression membranaire de cKit et favorisant la prolifération des progéniteurs granulocytes-macrophages (GMP) ainsi que la génération de granulocytes Gr-1⁺ [5].
In vivo, l'injection intrapéritonéale d'Inosinic acid (45mg/kg)administrée 15 minutes après une irradiation par rayon X à des souris mâles de sourche Kv:SHK soumises à une irridiation corporelle totale de 7 Gy, a significativement augmenté le taux de survie après cette dose létale. Le traitement a atténué la leucopénie et la thrombocytopéine radio-induites , tout en réduisant le niveau de dommages à ADN dans les cellules de la moelle osseuse et du thymus[6]. Dans un modèle de sepsis chez des souris C57BL/6J âgées de 10 à 12 semaines, l'injection intrapéritonéale d'Inosinic acid (1000mg/kg) a significativement inhibé la production de TNF-α et augmenté les taux d' IL-10[7].

Méthode de préparation | Les cellules Lineage-/low ont été isolées de la moelle osseuse de souris C57BL/6 de type sauvage à l'aide d'un kit de déplétion des cellules lignées. Les cellules ont été cultivées en milieu SFEM supplémenté avec 10ng/mL de facteur des cellules souches, 10ng/mL de thrombopoïétine et 10ng/mL d'IL-3, à 37°C et sous 5% de CO₂. Les cellules ont été traitées avec Inosinic acid à des concentrations de 0 à 15mM pendant 24-72 heures. |

Domaines d'application | Le traitement à Inosinic acid a significativement stimulé la prolifération des cellules progénitrices granulocytes-monocytes (GMP) et la production de granulocytes Gr-1+ de manière dose-dépendante. Inosinic acid a augmenté de 1.3 fois l'incorporation de BrdU dans les cellules GMP et accru de 1.4 fois la proportion de cellules Gr-1+ par rapport aux cellules témoins. |

Méthode de préparation | Les souris ont reçu une administration intrépéritonéale d' Inosinic acid (1000mg/kg) 30 minutes avant le challenge à LPS (10mg/kg, i.p.). Les échantillons de plasma et d'organes (rate, foie, poumon) ont été prélevés 90 minutes après LPS pour l'analyse des cytokines. Pour les modèles de sepsis batérien, Inosinic acid (1000mg/kg, i.p.) a été administré 1 heure avant la ligature et ponction coecale (CLP) ou l'infection bactérienne intranasale (Streptococcus pneumoniae et Escherichia coli). |
