NECA est un agoniste non-sélectif des récepteurs à l'adhénosine[1]. NECA régule le niveau intracellulaire d' AMP cyclique (cAMP) en activant les récepteurs à l'adhénosine, affectant ainsi de nombreux processus physiologiques cellulaires, tels que l'anti-inflammation, la vasodilatation, l'inhibition de l'activité neuronale, la cardioprotection et la régulation immunitaire[2-3].
Dans les cellules H9c2, NECA (0.1–10μM; 20min) a bloqué la phosphorylation de GSK-3β H2O2-induite et l'expression de GRP94[4]. Dans les cellules musculaires squelettiques, NECA (100nM, 10μM; 19h) module CPT1B, stimule LCAD et inhibe l'expression génique de mRNA d' ACCβ[5]. Dans les cellules HREC, NECA (10μM; 24-72h) induit une augmentation temporelle de la prolifération cellulaire[6].
Dans le modèle de rats polycythémiques post-hypoxiques, NECA (0.05, 0.1µmol/kg; iv; injection unique) augmente significativement le niveau sérique d'érythropoïétine chez les rats atteints d'érythrocytose hypoxique [7]. Dans les rats qui n'ont été que vaccinés, l'administration intranasale de NECA (50μL; 1mM; administration intranasale; administration unique) provoque une augmentation significative du nombre total de cellules (TCC) dans le lavage bronchoalvéloaire, des neutrophiles et des éosinophiles[8].

Méthode de préparation | Les cellules ont été lavées deux fois avec une saline tamponnée au phosphate (PBS), puis incubées dans une solution de Tyrode pendant 2 heures avant les expériences. Pour examiner l'effet de NECA sur le potentiel de la membrane mitochondriale, la phosphorylation de GSK-3β et GRP94, les cellules ont été exposées à 800µM de H2O2 pendant 20 minutes pour provoquer des lésions oxydantes. Une gamme de concentrations de NECA (0.1–10μM) a été administrée 10 minutes avant l'exposition à H2O2. Dans l'étude explorant les rôles d' ERS (stress du réticulum endoplasmique) et de la voie de signalisation cGMP/PKG dans la cardioprotection NECA-induite contre les lésions oxydatives, les cellules ont été exposées à 0,1µM de NECA/20mM de KT5823 pendant 20min. 2-DG et KT5823 ont été appliqués pendant 10 minutes avant que les cellules ne soient exposées à NECA. |

Méthode de préparation | Cette expérience in vivo a utilisé le modèle de rats polycythémiques post-hypoxiques (EHPCM), qui induit une érythrocytose en exposant les rats à un environnement hypoxique à basse pression pendant une longue durée. Du 3ème au 6ème jour après la fin du traitement hypoxique, NECA a été administré par voie intraveineuse ou intra-péritonéale, et enfin une stimulation hypoxique à court terme a été administrée à nouveau. Le sang, les reins et les foies des rats ont été ensuite prélevés, et la teneur en Epo (érythropoïétine) dans le sérum et les tissus a été détectée pour évaluer l'effet régulateur in vivo de NECA sur la production d' Epo. |
