SKLB-03220 is an orally bioavailable covalent inhibitor targeting the histone methyltransferase EZH2, an established oncology drug target involved in epigenetic regulation. SKLB-03220 is structurally based on the clinically approved EZH2 inhibitor tazemetostat and demonstrates the ability to abolish epigenetic H3K27me3 marks, thereby altering chromatin state and transcriptional activity.
