Product Introduction
Suitable for animal tissues (heart, liver, kidney, muscle, testes, brain, spleen, etc.), cultured cells, RNA viruses, fruit flies, bacteria, white blood cells, whole blood, some plant tissues, and more. RNA Stabilization Solution is an aqueous, non-toxic tissue preservation fluid that can rapidly permeate the cytoplasm of fresh tissue cells. It stabilizes and protects intracellular RNA in situ without freezing. After removing tissue slices and immediately immersing them in RNA Stabilization Solution, the future extraction of RNA's quality and quantity is unaffected. The RNA Stabilization Solution eliminates the inconvenience of needing to process RNA samples immediately or store them in liquid nitrogen. After immersion in RNA Stabilization Solution, RNA in fresh tissue cells can be perfectly preserved at 37˚C for a day, at 25˚C for a week, at 4˚C for a month, and for long-term storage at -20˚C or -80˚C. RNA virus samples (such as HCV and HIV) can be stored at 37˚C for a month. In addition to preserving RNA, this product can also preserve DNA and proteins.
Usage Instructions
RNA Stabilization Solution is only intended for use with fresh tissues. Do not freeze the tissue before immersing it in RNA Stabilization Solution! Simply cut the fresh tissue into pieces with a length, width, or height of less than 0.5 cm and immerse the pieces in RNA Stabilization Solution (RNA Stabilization Solution has strong penetration ability, so a thickness of one side should not exceed 0.5 cm). Immerse the fresh tissue in RNA Stabilization Solution at a 5-fold volume ratio, and store it at the appropriate temperature as instructed.
1.Animal Tissues: RNA Stabilization Solution does not disrupt or dissolve tissue structure. Therefore, tissues that have reached osmotic equilibrium by soaking in RNA Stabilization Solution can be taken out, further sectioned into smaller pieces, and then returned to RNA Stabilization Solution for future use. Small organs such as mouse liver, kidney, and spleen can be stored intact in RNA Stabilization Solution without the need for sectioning.
2.Plant Tissues: Cut plant tissues into sizes smaller than 0.5 cm and immerse them in RNA Stabilization Solution. Some plants have natural barriers like waxy layers that need to be disrupted to facilitate the penetration of RNA Stabilization Solution.
3.Tissue Culture Cells: After detaching cells, collect them by centrifugation, discard the supernatant, wash once with ice-cold PBS buffer to remove residual culture medium. Suspend cells in a small amount of PBS buffer, add 5-10 times the volume of RNA Stabilization Solution, and mix thoroughly.
4.Blood and Plasma: Red blood cells and white cells separated from serum can be stored using the same method as tissue culture cells. RNA Stabilization Solution can also be used to preserve whole blood, serum, and plasma. For whole blood, add 3 times the volume of RNA Stabilization Solution, and mix thoroughly.
5.Yeast: Collect 3x108 cells by centrifugation (>12,000g for 2 minutes). Immediately resuspend the cell pellet in 0.5-1 ml of RNA Stabilization Solution. Yeast cells can be stored in RNA Stabilization Solution at 25˚C for 8 hours, or at 4˚C for one week. For longer storage, after placing the yeast cell pellet in RNA Stabilization Solution for one hour, centrifuge again at >12,000g for 5 minutes, place the yeast cell pellet in liquid nitrogen, flash-freeze, and store at -80˚C.
6.Bacteria: RNA Stabilization Solution does not disrupt bacteria, but bacteria cannot grow in RNA Stabilization Solution. Bacteria can be preserved in RNA Stabilization Solution for up to one month while still allowing full RNA extraction.
Sample Preservation in RNA Stabilization Solution
1.Stored at -80˚C:
This method allows for long-term preservation of samples. Place the samples in RNA Stabilization Solution directly into -80˚C. When ready to use, samples can be thawed at room temperature and can also be refrozen without affecting the integrity and yield of the RNA.
2.Stored at -20˚C:
Place the samples in RNA Stabilization Solution into -20˚C. Samples won't freeze but might form some crystals; this will not impact future RNA extraction. Samples can be thawed at room temperature when needed and refrozen without compromising the RNA's integrity and yield.
3.Stored at 4˚C:
Samples can be stored at 4˚C for up to a month.
4.Stored at 25˚C:
When stored at 25˚C, the RNA in samples remains intact for one week. Samples stored for 2 weeks show slight RNA degradation. They can just about be used for Northern analysis, but the quality is sufficient for Nuclease Protection analysis or RT-PCR analysis.
5.Stored at 37˚C:
When stored at 37˚C, the RNA in samples remains intact for 24 hours, but there's some degradation after 3 days.
RNA Extraction from Samples in RNA Stabilization Solution
Remove the sample from the RNA Stabilization Solution. The RNA Stabilization Solution can be directly poured into the sink, and rinsed with tap water, no special treatment required.
1.Tissue:
Using clean tweezers, retrieve the sample from the RNA Stabilization Solution. Blot off any remaining RNA Stabilization Solution with absorbent paper. The tissue can then be processed similarly to fresh tissue, ground in liquid nitrogen, followed by homogenization. Then, proceed with the standard RNA extraction protocol.
2.Cells:
For cells stored in RNA Stabilization Solution, there are two options. One is to remove the RNA Stabilization Solution before extracting the RNA, and the other is to directly extract from the mixture of cells and RNA Stabilization Solution.
1) Remove the RNA Stabilization Solution before extracting RNA.
Cells stored in RNA Stabilization Solution can withstand high centrifugation speeds (about x5000g) without rupturing. Because the strength of each cell type varies, it's advisable to conduct preliminary experiments with less critical cells to ensure that the chosen speed does not damage them. Additionally, before centrifugation, one can dilute the mixture of RNA Stabilization Solution and cells with an equal volume of PBS to reduce the solution density and facilitate cell sedimentation.
2)Extract RNA directly without removing the RNA Stabilization Solution.
One can directly add 10 times the volume of a one-step RNA extraction reagent to the mixture of cells and RNA Stabilization Solution, and then extract RNA following standard procedures.
