Iristectorin BはIris tectorumに由来する天然のイソフラボンであり、抗癌作用を持つ[1]。Iristectorin Bは、リポ多糖(LPS)によるNOの産生を阻害することで、抗炎症活性を発揮できる[2]。Iristectorin Bは内部標準物質として広く使われ、関連する化合物の抽出と分離のためのイオン液体超音波併用抽出法を開発する[3]。
In vitro(体外)実験で、Iristectorin B(25µM)で24時間処理したところ、グルコース・酸素の欠乏/再酸素化(OGD/R)処理後、PC12細胞の生存率が著しく向上し、細胞アポトーシスが減少し、TFR1、HMOX1とSLC3A2の発現が著しく低下した[4]。

準備方法 | 37℃、5% CO2の存在下で、10%のウシ胎児血清(FBS)と1% のペニシリン/ストレプトマイシンを添加した高グルコースDMEM培地でPC12細胞を培養した。10mM Na2S2O4を含有する100μlの無糖EBSS溶液を添加し、2時間インキュベートした。再酸素化のために、細胞を正常な培地に替え、引き続き24時間培養し、虚血脳損傷を模倣したOGD/R損傷モデルを構築した。OGD/R処理後、6.25µM、12.5µMと25µMのIristectorin Bを含有する正常培地で24時間添加、培養した。細胞の生存率を分析した。 | ||
