Phenelzineはモノアミン酸化酵素(MAO)である[1]。Phenelzineは脳で、モノアミン酸化酵素AとBを不可逆的に阻害し、ノルエピネフリン、ドーパミン、セロトニンなどの神経伝達物質の分解を減らし、よってそれらのレベルを増やし、気分を改善する[2-3]。Phenelzineは主に、難治性鬱病の治療に使われる[4]。
PC12細胞で、Phenelzine (1-50μM;24時間)は、1-メチル-4-フェニルピリジニウム(MPP+)による細胞核の凝縮と断片化を減らした[5]。3T3-F442Aと3T3-L1細胞で、タンパク質-1c (SREBP-1c)と結合する細胞トリグリセリドおよびステロール調節要素の減少は、より低い濃度のPhenelzine (1-10μM;8日間)で検出された[6]。Molt-4細胞で、Phenelzine (5-25μM;24時間)はヒストンH3K4のモノメチル化、H3K4ジメチル化、ヒストンH3アセチル化のレベルを上方制御し、p21とp15タンパク質の発見を上方制御し、DNMT1の発見を下方制御し、最終的にMolt-4細胞の増殖を阻害し、細胞のアポトーシスを誘発した[7]。
実験性自己免疫脳脊髄炎(EAE)マウスモデルで、Phenelzine (15mg/kg;腹腔内注射;28日間)は発症を遅延させ、EAEマウスの慢性期を軽減し、探索行動や情緒障害などの非運動症状を著しく改善できる[8]。実験性自己免疫脳脊髄炎マウスモデルで、Phenelzine (15mg/kg;腹腔内注射;14日間)は中枢神経系で、GABA、ドーパミンと生物アミノのレベルを上昇させる[9]。

準備方法 | 急性phenelzine処理実験で、研究者は通常、実験性自己免疫脳脊髄炎(EAE)マウスに対しては疾患のピーク(スコア≥ 3)で、あるいは対照のマウスに対し、30mg/kgのPhenelzine(遊離塩基として抗菌水に溶解)を単回腹腔内注射する。投与の3時間後に、マウスを殺処分し、中枢神経系組織を除去し、後続する神経伝達物質分析のために冷凍保存した。慢性処理実験については、モデルを作成した後の7日目から、28日間連続で、15mg/kgのPhenelzineまたは同量の溶媒を毎日腹腔内注射した。免疫組織化学またはHPLC分析のために、処理の翌朝に、中枢神経系(CNS)の組織を採取した。 |
