Phlorofucofuroeckol A is a FcεRI inhibitor with low cytotoxicity toward basophilic leukemia cells. Phlorofucofuroeckol A suppresses binding activity between IgE and FcεRI receptor. Phlorofucofuroeckol A inhibits histamine release from basophilic leukemia cells stimulated by IgE/anti-IgE or calcium ionophore A23187. Phlorofucofuroeckol A inhibits β-hexosaminidase release from basophilic leukemia cells stimulated by IgE/DNP-BSA. Phlorofucofuroeckol A can be used for the research of allergy disease [1].
In Vitro, Phlorofucofuroeckol A (10-100 μM; 48 h) shows no significant cytotoxicity toward KU812 and RBL-2H3 cells[1]. Phlorofucofuroeckol A (10-100 μM; 30 min) dose-dependently inhibits IgE/anti-IgE-induced histamine release in KU812 cells, with an IC50 of 65.81 μM, and reduces relative histamine release to 34.54% at 100 μM after pre-incubation[1]. Phlorofucofuroeckol A (10-100 μM; 30 min) dose-dependently inhibits A23187-induced histamine release in RBL-2H3 cells, with an IC50 of 64.06 μM, and reduces relative histamine release to 34.18% at 100 μM after pre-incubation[1]. Phlorofucofuroeckol A (10-100 μM) dose-dependently inhibits IgE/DNP-BSA-induced β-hexosaminidase release in RBL-2H3 cells, with an IC50 of 38.87 μM, and reduces relative β-hexosaminidase release to 20.80% at 100 μM[1]. Phlorofucofuroeckol A (12.5-50 μM) dose-dependently inhibits IgE-FcεRI binding on KU812F cells, reducing the percentage of positive cells to 79.51%, 57.58%, and 34.23% at 12.5, 25, and 50 μM, respectively[1].
