Duvortuxizumab (MGD 011) is a chimeric humanized IG antibody targeting CD19 and CD3E [1].
In Vivo, Duvortuxizumab (0.16-100 μg/kg, iv, once every 3-4 days for 8 doses) exhibits antitumor activity in HBL-2 xenograft mouse models[2].
In Vitro, Duvortuxizumab is a dual-affinity re-targeting (DART) protein that binds to the CD3 molecule on T cells and the CD19 antigen on B cells. Duvortuxizumab exhibits good affinity to human CD3 and human CD19 with KDs of 21.2 nM and 2.0 nM[2]. Duvortuxizumab exhibits cytotoxicity to CD19-positive cell Raji/GF (EC50 is 0.02-0.03 ng/mL), causes depletion of malignant B cells in human PBMCs (EC50 is 0.018 pg/mL)[2].
