Description of RBITC

RBITC,o isotiocianato de Rodamina B,es un colorante fluorescente conocido por su grupo isotiocianato que puede unirse covalentemente a aminas primarias en proteínas y biomoléculas[1][2]. RBITC tiene una longitud de onda máxima de excitación de 562 nm y una longitud de onda de emisión de 582 nm[3]. RBITC se utiliza principalmente en biología celular[4],biología molecualr e inmunología para la localización de proteínas[5], el seguimiento celular[6],y el estudio de procesos celulares[7].

Protocol of RBITC

Protoclo experimental para el marcaje de proteínas con RBITC:

1.Preparación de la solución RBITX: Disolver RBITC a 10mg/mL en DMSO antes de su uso.Homogeneizar.

2.Preparación de la solución proteica: Disolver la proteína objetivo en fosfato de sodio 0,1M,0,15M tampón NaCI a pH 7,2 para fomar una solución proteica d e1 mg/mL.

3.Reacción de marcado:Transfiere la solución de RBITC a la solución proteica en una proporción molar de 5:1 y remueve suavemente la mazcla en la pscuridad a 4ºC durante 12 horas.

4. Diálisis:Eliminar RBITC no reaccionado mediante diálisis o con una columna de eliminación de tintes.

5.Tras la diálisis,recoger la solución proteica marcada,determinar su concentración y almancenar la prote´´ina marcada con RBITC a 4ºC protegida de la luz hasta que esté lista para su uso.

Este protocolo solo proporciona una guía y debe modificarse según tus necesidades específicas.
