Description of Sedanolide

Sedanolide, un tipo de ftaleína, es un potente inhibidor de hMAO-B con un valor de IC50 de 0.124±0.033µM [1]. Sedanolide tiene actividades inhibidores de la COX-I y la COX-II y actividades inhibidoras de la topoisomerasa-I y topoisomerase-II[2]. Sedanolide se ha usado en una variedad de modelos de células cancerosas para inhibir el crecimiento celular y tiene efectos inhibidores en una variedad de bacterias y hongos[3].

In vitro, el tratamiento con Sedanolide a 500μM durante 24 horas indujo la autofagia en las células J5, que inhibieron la vía PI3K/Akt/mTOR[4]. El tratamiento con Sedanolide 500μM durante 24h resultó en una aumento significativo en las roturas de la cadena de DNA en las células HepG2[5]. Tratamiento con Sedanolide 100μM para las células HepG2 que expresan luciferasa protegidas durante 24h de la muerte celular inducida por H2O2 y los niveles reducidos de ROS intracelular[6].

In vivo, el tratamiento con Sedanolide a través de la administración diaria a una dosis de 20mg/kg/día durante 14 días consecutivos protegió a los ratones de la colitis inducida por sulfato de sodio de dextrano (DSS), alivió la inflamación intestinal y mejoró la función de barrera intestinal[7]. La administración oral de Sedanolide (10mg/kg/día) durante 14 días consecutivos puede activar la vía Nrf2 y aliviar el daño hepático en ratones tratados con Acetaminophen (APAP)[8].

Células HepG2 que expresan luciferasa

Método de preparación

Las células de HepG2 que expresan luciferasa se cultivaron a 37°C y al 5% de CO2 en un ambiente humidificado en el medio Eagle modificado de, complementado con un 10% de suero bovino fetal (FBS), un 1% de aminoácidos no esenciales (MEM) de medio esencial mínimo y piruvato de sodio de 1mM. Las células HepG2 que expresan luciferasa se sembraron en placas de 12 pozos a una densidad de 3.6×105 células/pocillo. Después de un día de incubación, las células fueron pretratadas con Sedanolide 100µM durante 24h y luego expuestas a peróxido de hidrógeno durante 24h adicionales antes del análsis de la muerte celular inducida por peróxido de hidrógeno.

Modelos animales

Ratones macho C57BL/6J

Método de preparación

Los ratones machos C57BL/6J de seis semanas de edad fueron criados en un entorno SPF y se les proporcinó agua y alimentos adecuados. Los ratones fueron expuestos a un ciclo de luz/12 hoiras y oscuridad de 12 y se mantuvieron a una temperatura de 23±2°C. Se dividieron aleatoriamente en el grupo NC y el grupo Sedanolide. Los ratones fueron avavados por vía oral con Sedanolide (20mg/kg) o el vehículo todos los días durante 14 días consecutivos, desde el séptimo día hasta el día 14. En el grupo Sedanolide, se administró un 2% de DSS a los ratones del séptimo al decimocuarto día. El día 15th, el agua fue sustituida por un 2% de DSS, y los ratones fueron anestesiados y sacrificados el día 16th. Los tejidos del colon fueron recogidos para su análisis.

Áreas de aplicación

El tratamiento con Sedanolide mejoró los síntomas de la colitis, los niveles de inflamación y la función de la barrera intestinal en ratones.
