Oleic acid (9-cis-Octadecenoic acid), a major monounsaturated fatty acid (MUFA) in the circulatory system, is an energy molecule and an element of cell membranes[1]. Oleic acid diminishes the expression of cholesterol transport-related proteins, decreases cholesterol absorption, and reduces the oxidation of low-density lipoprotein (LDL), preventing atherosclerosis[2]. Oleic acid has been widely used in diabetes research to inhibit endoplasmic reticulum (ER) stress, prevent the weakening of the insulin signaling pathway, and improve the survival of beta cells[3].
In vitro, Oleic acid treatment for 72 hours significantly reduced the proliferation of KLE, Hec-1B and ECC-1 cells, with IC50 values of 445.6μM, 382.8μM and 369.8μM, respectively[4]. Treatment with 25μM Oleic acid for 48 hours inhibited stearic acid-induced growth inhibition and pro-inflammatory responses in human aortic endothelial cells (HAECs)[5]. Treatment with 1.32mM oleic acid for 24 hours induced lipid accumulation and increased the expression of PPARγ and SREBP-1 in HepG2 cells[6].
In vivo, Oleic acid treatment via a single intratracheal injection at a dose of 10mg/kg within 24 hours elicited pulmonary inflammation in Swiss Webster mice, promoting the activation of lung white blood cells and the production of inflammatory mediators[7]. Oral administration of Oleic acid (160mg/kg) daily for 14 days improved the Angiotensin II-induced cardiac remodeling, and inhibited cardiomyocyte hypertrophy and collagen deposition in mice[8].

Preparation Method |
Male C57BL/6 mice (10-week-old) were housed in a specific-pathogen-free environment under a 12h/12h light-dark cycle and fed rodent diet ad libitum. After mice orally received Oleic acid (160mg/kg/day) or saline for 14 days, cardiac remodeling was induced by chronic infusion of Ang II at a dose of 1000ng/kg per minute for 2 weeks with a pump. Mice were anesthetized with 3.0% isoflurane mixed with oxygen (100%; airflow velocity: 1L/minute) was used to maintain anesthesia. An incision was made in the skin on the back of the mouse, and the pump was implanted into the subcutaneous tissue. After being sutured and regaining consciousness, the mice were returned to their original cages and fed individually until the end of the experiment. Heart was excised and prepared for histology. |
