PPACK ist ein potenter, selektiver und irreversibler Thrombininhibitor, der humanes Alpha-Thrombin mit einem Ki-Wert von 0.24 nM hemmt. PPACK hemmt die Bindung von rt-PA an Plasmaproteaseinhibitoren in vitro[1-3]. Es bildet eine kovalente Bindung an das Ser im aktiven Zentrum von Thrombin und vernetzt sich dort mit His57 zu einem tetraedrischen PPack-Thrombin-Komplex. PPACK hemmte Veränderungen der Fibrinabbauprodukte, des Plasminogens und des Alpha-2-Antiplasmins vollständig[4-6].
Koinkubation mit PPACK(0.2 mM; 2 Stunden) reduzierte den durch Plasmin induzierten Anstieg der Endothelpermeabilität in bovinen Aortenendothelzellen um 59%[7]. Hämoxygenase(HO)-1 ist ein stressinduzierbares, geschwindigkeitsbestimmendes Enzym im Häm-Abbau, das Zytoprotektion gegen oxidative Schäden bietet. Die Vorbehandlung von Zellen mit PPACK(24h; 30 nM) antagonisierte wirksam den verstärkenden Effekt von Thrombin auf die HO-1-Expression[8].
Eine Vorbehandlung mit PPACK(Bolusdosis von 10-100 μg/kg oder Infusion von 0.25-1.0 μg/kg/min) kann Thrombin-induzierte zerebrale Thromboembolie bei Kaninchen hemmen[9].

Kinetische Verfahren(Inhibition von Thrombin mit PPACK) [1]: | |
Vorbereitungsmethode | Die Thrombin-Stammlösung wurde im entsprechenden Puffer verdünnt. Dadurch entstand eine 20 mL Lösung mit einer Konzentration von 3-5×10-10 M. Ein Aliquot von 980 μL wurde in einer Küvette im temperaturgeregelten Zellkompartiment des Spektrophotometers inkubiert, um ein thermisches Gleichgewicht zu erreichen. Die Thrombinhemmung wurde durch Injektion von 10 μL einer thermisch äquilibrierten 1.1 × 10-7 to 3.65 × 10-7 M PPACK-Lösung und Mischen eingeleitet. Das Reaktionsgemisch wurde sofort für einen Zeitraum zwischen 15 und 200 Sekunden inkubiert. Danach wurden schnell 10 μL einer 3-5×10-3 M Substratlösung in DMSO hinzugefügt, um die verbleibende Enzymaktivität zu messen. Geschwindigkeitskonstanten Pseudo-erster Ordnung wurden berechnet, dadurch dass die Exponentialfunktion an die verbleibende Enzymaktivität gegenüber die Hemmzeit für 10 aufeinanderfolgende Inkubationszeiten angepasst wurde. Die Thrombinaktivität blieb während der gesamten Dauer des Experiments im Temperaturbereich von 288-308 K konstant. |

Vorbereitungsmethode | Drei bis fünf Tage nach der Plattierung auf Polycarbonat-Membranclusterplatten wurden konfluente Monoschichten der Rinderaorta-Endothelzellen gewonnen. Die Zellen wurden mit Plasminogen und einem Plasminogenaktivator behandelt. PPACK wurde in die oberen Kompartimente gegeben. Die Monoschichten wurden dann zwei Stunden lang in einer 5% CO2 / 95% O2 Atmosphäre inkubiert. |

Kaninchen wurden 1 Minute vor oder nach der Verabreichung von Thrombin mit einem PPACK-Bolus behandelt oder erhielten 30 Minuten vor der Verabreichung von Thrombin eine Infusion mit PPACK, die eine Stunde lang mit einer Rate von 0.1 mL/min fortgesetzt wurde. |
