Description

Albiglutide Fragment is the active fragment (7-36) of Albiglutide, a glucagon-like peptide-1 (GLP-1) mimetic belonging to the class of long-acting GLP-1 receptor agonists. Albiglutide Fragment can be used in research related to type 2 diabetes (T2D)[1-4].

In vitro, RIN 1046-38 cells were co-treated with Albiglutide Fragment (10nM) and glucose (5mM) for 0.5-1 hours. Albiglutide Fragment significantly increased insulin secretion[5]. CA-77 cells were incubated with Albiglutide Fragment (0.01-10nM) in the presence of 2mM calcium for 2 hours. Albiglutide Fragment dose-dependently stimulated calcitonin secretion[6].

In vivo, traumatic brain injury (TBI) model rats received daily intracerebroventricular (i.c.v.) injections of Albiglutide Fragment (1nmol or 10nmol in 5μl) for 30 days. Albiglutide Fragment significantly improved neurological dysfunction and cognitive deficits after TBI[7]. NMRI mice received a single intravenous injection of Albiglutide Fragment (32nmol/kg). Albiglutide Fragment significantly reduced plasma glucose levels and increased plasma insulin levels in mice. When Albiglutide Fragment was combined with 2,5-AM, an additive hypoglycemic effect was produced[8].

Protocole (Protocol)

Cell experiment [1]:

Cell lines

RIN 1046-38 cells (rat insulinoma cell line) and CHO/GLP-1R cells (Chinese hamster ovary cells stably transfected with the human GLP-1 receptor)

Preparation Method

RIN 1046-38 cells were routinely cultured in M199 with Earle's salts supplemented with glucose (11mM), penicillin (50U/ml), streptomycin (50ug/ml), and glutamine (2mM) in a humidified 5% CO₂-95% air incubator at 37°C. CHO/GLP-1R cells were grown to confluency on plates. For insulin secretion assay, RIN 1046-38 cells were incubated with Albiglutide Fragment (10nM) and glucose (5mM) for 1h at 37°C. For cAMP determination, RIN 1046-38 cells were incubated with Albiglutide Fragment (10nM) and IBMX (1mM) for 0.5h at 37°C.

Reaction Conditions

10nM; 0.5-1h

Applications

Albiglutide Fragment significantly increased insulin secretion from RIN 1046-38 cells, approximately 2-fold above basal levels. In cAMP assays using RIN 1046-38 cells, Albiglutide Fragment induced intracellular cAMP production.

Animal experiment [2]:

Animal models

Male Sprague-Dawley (SD) rats (10-12 weeks old, weighing approximately 250g) subjected to traumatic brain injury (TBI)

Preparation Method

A severe TBI model was established via a controlled cortical impact. Immediately after TBI, rats received Albiglutide Fragment (1nmol or 10nmol in 5μl) intervention via intracerebroventricular (i.c.v.) injection once a day for 30 days.

Dosage form

1nmol or 10nmol in 5μl; i.c.v.; once daily for 30 days

Applications

Albiglutide Fragment significantly improved neurological dysfunction and cognitive deficits, reduced cerebral edema, decreased the levels of H₂O₂ and intracellular ROS, increased cellular antioxidant factors (CAT, SOD, GSH), promoted the activation of the ERK5/CREB signaling pathways, and suppressed apoptosis (reduced CC-3 and Bax/Bcl-2 expression) in the hippocampal tissues of TBI rats.
