Concanavalin ACon A, derived from Canavalia ensiformis beans, is a lectin protein (Mw 104 kDa). Under the condition of p27.0, Con A exists in a homogenous tetrameric structure, consisting of four subunits with a molecular weight of 26 kDa. Under acidic conditions (pH 4.5-5.5), on A dissociates into an activated dimeric structure of 52 kDa. Each monomer, regardless of pH or molecular structure, contains two metal case binding sites. Metal nanoparticles C82+and M2+must bind to both sites in order for Cn A to be active. Awakening is the process of generating a stable form of dimeric structure from other living organisms.
Concanavalin A (Con A) has no blood type specificity and selectively binds to - mannose/residues and a-glucose residues on various proteins, sugars, and class molecules. It requires the presence of Ca2+and Mn2+ions to display its activity. Con A is suitable for carbohydrate research, glycoprotein purification, enzyme labeling, thin membrane studies, Shao cell agglutination, and cell typing studies.
This is Alexa Fluor 594 labeled Concanavalin A (Alexa Fluor 594 ConA), purified by affinity purification at Ex/Em=590/617nm. The recommended working concentration range is 5-25 μ G/ml.
Operation process
1、 Preparation of storage solution
Before use, place Alexa Fluor 594-ConA (1mg) stored at low temperature for 30 minutes at room temperature, leave at low speed for 2-3 minutes, and add 500ul ddH2O (sterile) to prepare a 2mg/ml storage solution. Short term use can be stored at 2-8 ° C, and long-term use should be packaged and stored at -20 ° C according to a single dose to avoid repeated freeze-thaw cycles. It should be stable for at least one month.
2、 Live cell staining
[Note 1] Before use, place the low-temperature stored product at room temperature to completely melt it, then centrifuge at low speed and extract the supernatant for subsequent experiments. This step can remove protein aggregates that may form during the storage process of the product, thereby reducing non-specific background staining.
[Note 2] Cona protein conjugates (such as Alexa Fluor 594 ConA) stain the cytoplasmic membrane and ultimately stain the intracellular vesicles after invagination of the membrane
3、 Fixed cell staining
[Note 1] Before use, place the low-temperature stored product at room temperature to completely melt it, then centrifuge at low speed and absorb the supernatant for subsequent experiments. This step can remove protein aggregates that may form during the storage process of this product, thereby reducing non-specific background staining.
[Note 2] Cona protein conjugates (such as Alexa Fuor 594 ConA) do not exhibit membrane permeability, therefore, fixed (non permeable) cells exhibit cytoplasmic membrane staining. If fine fixed and then subjected to chemical treatment, it may cause staining of the cytoplasmic membrane.
matters needing attention
1) Long term storage of this product may result in precipitation. Please incubate at 37 ℃ for a few minutes before use, and then centrifuge and extract the supernatant for use. This operation will not have a negative impact on performance.
2) Fluorescent dyes all have quenching problems, so be careful to avoid light during storage and operation.
